Objective To standardize the evaluation system of the quality standard of MAY medicinal slices.To research the inhibition of petroleum ether layer of MAY on Human tumor cells.To study the effects of petroleum ether layer of MAY on S180 and H22 of tumor-plant mice.To provide experimental basis for the research and development of new drugs of traditional Chinese medicine.Methods(1)The original plant configuration,character,organization character,powder character and physicochemical property of MAY were identified.Oven-drying method,Ash determination method and hot-dip method,recorded in "Chinese Pharmacopoeia"(2015 version),were employed to determine the contents of water,total ash,ethanol-soluble extractions and water-soluble extractions of MAY.Physicochemical identified method was employed to determine the ingredients of MAY.The method of phenol-concentrated sulphuric acid was employed to determine the content of polysaccharide of MAY.The method of PMP was employed to determine the constitutes and its contents of polysaccharide of MAY(2)CCK-8 was employed to determine the inhibitions of petroleum ether layer of MAY on A549,PC-3,HepG2,MCF-7,231-LUC,Bel-7402,MGC-803,SGC-7901(100,200,300,400,500,600,700,800 μg/mL),A375,HT-29(100,200,400,600,800,1000,1200,1500μg/mL).The IC50 of these cells were:A549:364.0μg/mL,95%[249.5,458.6]、PC-3:156.0μg/mL,95%[91.54,209.7]、HepG2:347.0μg/mL,95%[243.8,445.1]、MCF-7:276.0μg/mL,95%[215.2,332.2]、231-LUC:315.8μg/mL,95%[167.9,468.2]、Bel-7402:353.0μg/mL,95%[45.7,588.8]、MGC-803:500.0μg/mL,95%[444.1,548.8]、SGC-7901:490.0μg/mL,95%[472.7,507.0]、A375:696.1μg/mL,95%[506.5,901.2]、HT-29:629.8μg/mL,95%[453.1,771.5].(3)The tumor-plant mice models were randomly divided into 5 groups:Model group,petroleum ether layer of MAY(PELA)groups(0.4、0.8、1.6g/kg),cisplatin group(S180:5mg/kg,H22:2mg/kg).The mice were given with the corresponding dosage of drugs(0.1mL/10g)by lavage for 10 days.After the last drug,the mice could be pasted but water for 12h.The mice were sacrificed with the tumors,thymus and spleens peeled off and weighed.The efficacy of petroleum ether layer of MAY was determined by tumor inhibition rate,indexes of thymus and spleen,HE staining results of tumors.Results(1)The morphology,character of Traditional Chinese Medicine and microscopic character of MAY were identified,and the features identification were described.Physical and chemical test:The main ingredients of MAY were phenols,tannins,saponins,organic acids,flavonoids,anthraquinone,sterol,triterpene,alkaloids.Examination was measured in nine MAY samples of moisture content(9.72、7.85、9.64、7.85、7.47、7.86、7.47、7.41、9.71)%,total ash content(10.28、9.17、10.61、9.34、10.77、10.85、10.77、9.19、10.27)%,water-soluble extractions contents(11.11、10.56、10.76、9.18、10.15、10.47、9.60、11.81、9.63)%and ethanol-soluble extraction(15.78、21.09、16.87、16.15、18.96、16.19、20.90、16.98、20.85)%,polysaccharide content(8.08、8.17、8.13、8.99、8.46、10.01、10.58、9.17、9.66)%.The contents of polysaccharide were mannose(4.22%),galacturonic acid(7.10%),glucose(77.06%),galactose(9.83%),arabinose(1.80%).(2)All of the ten human tumor cells could be inhibited by petroleum ether layer of MAY.PC-3 could be furthest inhibited and its IC50 was 156.0μg/mL.(3)Compared with model group,the tumor inhibition rates of PELA on S180 were 29.05%、39.88%、40.56%respectively(P<0.01).And the rats of PELA on H22 were 20.38%、32.18%(P<0.01)、46.67%(P<0.01)respectively.There was no significant differences between S180-plant mice,H22-plant mice and model mice on indexes of thymus and spleen and was improved on the results of tumors HE staining of S180-plant and H22-plant mice.Conclusions(1)Here were obvious identified characters on morphology,character of Traditional Chinese Medicine and microscopic character of MAY,which had provided Pharmacognosy identification basis for systemic identifi cation of MAY.A systemic methods of quality study had established by physical and chemical test,water content test,total ash content test,water-soluble extractions and ethanol-soluble extraction tests and polysaccharide content text,polysaccharide content text.(2)All of the ten human tumor cells could be inhibited by petroleum ether layer of MAY.PC-3 could be furthest inhibited and its IC50 was 156.0μg/mL.(3)PELA(0.4、0.8、1.6g/kg)could significantly inhibited S180 and H22 without any obvious toxic and side effect on immune organs of mice. |
