A Preclinical Study Of Related Mechanism Between Hippocampal & Immunocyte Cell Apoptosis And Chronic Psychological Stress In Mice With S₁₈₀ Tumor

Objective: This research mainly aimed to observes the effect of restraint stress on neuron in the hippocampus ,which is the variety of the number of neuron and their apoptosis; Observe the morphological change of adrenal cortex after stress ;and to explore the apoptosis of central and periphery immune organ thymus and spleen following restrain stress; while we measure the growth of tumor in mice bearing S180 cell after stress, Thus to look forward to inquiry into the variety of endocrine immunity function and that of hippocampus, and subsequently to discover the possible mechanism between hippocampus damage and restraint stress in the member level, while to elaborate the possible mechanism between immunity function deregulation and stress .Then we expect to furtherly Explore the related mechanism that the psychological stress influence on tumors occurrence and development.Methods:Sixty four male Kunming mice were employed as experimental animal model,Mice were randomly divided into four groups : normal control group(NC),simple stress group(SS),simple tumor group(ST),tumor with restrain stress group(S+T)。Tumor cells were extracted from abdominal cavity of mice with S180 tumor ,then were washed with 0.9%NS twice then diluted to 1x107/l with RPMI 1640 ,the diluted cells then were injected subcutaneously in the right oxter in 0.2ml/mouse,and restraint stress was imposed on mice for 8 hour per night for fourteen days。Fourteen days later mice were killed,thymus,spleen,tumor were taken out and weighted before put into 4% paraform ,and adrenal gland,brain were also put into 4%paraform for further study。Normally make the paraffin section and immunohistochemistry for Caspase-3,then count the positive cell with microscope and take photomicrographs,Utilizing t test to analyse the collected data in SSPS 11.5。Result:(1)Should arouse the small rat adrenal glands skin quality to increase obviously thick, tie a form to take cell to increase to living fatty big also take off grain, hint the sugar skin quality hormone to secrete increment, the establishment tie should arouse model success.(2)At the of test, the average...
