| China is the country of origin of litchi and the largest producer in the world.While it is planted on a large scale,a large number of branches produced by pruning have not been reasonably utilized.The disposal methods of burning,landfilling or random discarding will cause agricultural non-point source pollution,air pollution,fire and many other ecological problems,which is contrary to the concept of green development.Edible and medicinal fungi have the ability to decompose agricultural and forestry wastes,which is one of the effective means to solve non-point source pollution.The selection and breeding of good strains and the use of good methods are the most important innovation fields in the development of edible and medicinal fungi industry.Therefore,in this study,Pleurotus abalonus,a rare fungus,was used as the research object,and atmospheric plasma mutagenesis technology was used to breed excellent strains of P.abalonus which had the advantage of decompressing litchi branch debris,in order to realize the diversification of fungus cultivars.Maximize the utilization of forestry waste resources.In this paper,310 P.abalonus mutant strains were obtained through ARTP mutagenesis test.Through antagonistic test,comparison of growth rate and related enzymatic studies,it was determined that the characters of strains B75,B5 and B217 were better than the original strain,and strain B75 had the best characters.Compared with the starting strain,the mycelial growth rate was increased by 0.69 mm/d,the laccase activity was increased by 38.9%,and the cellulase activity was increased by 34.3%.ISSR molecular marker technology was used to determine the genetic similarity coefficient variation range between strains B75,B5,B217 and the starting strain,which was 0.433~0.720,indicating that there was a certain genetic distance between them,and the genetic distance between the strain B75 with the best character and the starting strain was the longest.Response surface methodology was used to optimize the liquid medium of B75,and the optimal formula was determined as glucose 2.20%,yeast extract 0.60%,soybean cake powder0.80%,corn powder 0.50%,potassium dihydrogen phosphate 0.10%,magnesium sulfate0.10%.In the research of the preparation process of liquid strain-grain original seed,comparing the effects of PDA strain,basic liquid strain and optimized liquid strain on the preparation of the original seed,it was found that the optimized liquid medium strain had more advantages in the preparation of liquid-grain original seed,and could effectively shorten the time of full tube.According to the growth rate and biological efficiency of mycelia,the optimum growth medium for P.abalonus B75 was 78% rice,20% litchi sawdust and 2% lime,The optimal culture medium formula was lychee sawdust 65%,corn cob 20%,bran 13%,lime 2%,by comparing the growth of strain B75 in different original culture medium formula and litchi sawdust culture medium formula.Under these conditions,the mycelia growth rate was 5.52mm/d,and the biological efficiency was 71.66%.By comparing the contents of crude polysaccharide,total flavonoids and ergosterol in the fruiting body of strain B75,B5,B217 and the original strain,it was found that the content of relevant active components in the fruiting body of strain B75 was the highest under the culture substrate of litchi branch debris,which again confirmed that strain B75 was the dominant strain.In the exploration of the circular economy model,the use of P.abalonus bran to grow Zisong No.3" found that a certain amount of P.abalonus bran is helpful for the growth of"Zisong No.3".And when the addition amount of fungus bran was 20%,the growth indexes of "Zisong No.3" were the best. |
