| The point of this research was the application of liquid strain of Pleurotus eryngii to solid plant. In this study, the culture medium of liquid strain was selected. The best suitable culture condition and culture period of liquid strain were determined. On the same time, in order to adapt the status quo that decentralization and bywork in miniature of edible fungus production in our country, the preservation method of the liquid strain of Pleurotus eryngii was also studied. So that the liquid strain can be easy to generalized. The main results were as follows:(1) The study was based on the indexes of the dry weight, the density and the diameter of the spawn pellet, and the spawn growing instance after inoculated it to the plate. The experiment proved that the liquid strain of the Pleurotus eryngii can growing best in the culture medium that use the soybean powder for the main material. The composing of the liquid culture medium as follow:soybean powder 6%, dextrose3%, peptone0.2%, KH2PO4 0.05%, MgSO40.05%, VB110mg/L.(2) The orthogonal testing method of L9(34) was taken to study the effect of different combination of liquid capacity, temperature, rev and inoculate quantity according to observing and measuring index of dry weight, density and diameter of Pleurotus eryngii spawn pellet.. The result showed that the optimum combination of the culture condition of Pleurotus eryngii is liquid capacity 100ml,temperature 30℃ ,rev 210r/min, inoculate quantity 15 %. These kind of liquid seeds of Pleurotus eryngii are suitable for planting. And the suitable culture period is 4 days.(3) The storage ways of the liquid strain of Pleurotus eryngii that the storage in the room temperature (1620℃), refrigerate in the refrigeratory (04℃), freeze in the ordinaire refrigeratory(-18-20℃) and deep freeze in the low temperature refrigeratory (-45℃) were studied. The result showed that the liquid strain of Pleurotus eryngii can be stored in the low temperature refrigeratory(-45℃), and the period of preservation can reached 90 days. In view of the cost, it can be refrigerated in the refrigeratory (04℃), but the period of preservation shouldn't excess 30 days. The period of preservation is 10 days in the room temperature (1620℃). It is unsuitable for the liquid strain of Pleurotus eryngii to freeze in the ordinaire refrigeratory (-18-20℃ ). |
