Research purposes: To establish an acute soft tissue injury model in SD rats by heavy impact method,observe and record the symptom index of affected limb injury and muscle morphological changes in rats at different time points after medication,and detect the contents of NO,SOD and MDA in the injured tissue To explore the efficacy and possible therapeutic mechanism of Jiawei No.2 Huoluo ointment in the treatment of acute soft tissue injury in rats.Research methods: 96 SD male rats(6 spares)were selected,and the rat acute soft tissue injury model was constructed by the heavy object blow method.The 72 successfully modeled rats were randomly divided into four large groups(n=18).,namely Group A(Jiawei No.2Huoluo Ointment Group),Group B(Erhuang Xinshang Zhitong Ointment Group),Group C(Voltarin Group),Group D(Model Control Group),18 unmodeled rats were used as Group E(Blank control group).Then five groups of rats were randomly divided into three groups(n=6)according to the three time points of day 1,day 4 and day 7 respectively.The corresponding drugs were externally applied to the model rats in groups A,B,C,and D on a regular and regular basis,and group E was not used for modeling.On the 1st,4th and 7th days after administration,the rats were killed in batches and the materials were collected.The symptom scores of the affected limbs,the morphological changes of the muscles and the contents of NO,SOD and MDA in the injured tissues were observed and recorded.Research results:(1)Soft tissue injury symptom score: There were significant differences between the four groups of model rats compared with the E group(P<0.05),indicating that the model was successful.There were significant differences between the three groups and the D group on the 4th and 7th day(P<0.05),indicating that the three groups of drugs can significantly improve the symptoms of subcutaneous blood stasis,limb swelling,and movement disorders in rats.There was no significant difference in the symptom scores on the 1st,4th,and 7th days among the three drug groups(P>0.05).(2)Histomorphological observation: Compared with group D,group A,B,and C could significantly improve the inflammatory response of local soft tissue in the affected limb of rats with acute soft tissue injury under light microscope observation,and promote the repair of damaged tissue.(3)NO content in injured tissue homogenate: compared within the group,the NO content in each modeling group showed a trend of first increasing and then decreasing,reaching a peak on the4 th day,and compared with the E group,there was a significant difference(P<0.05).Compared with the D group,the NO content of the three groups was lower than that of the D group on the 1st,4th and 7th days,and there was a significant difference(P<0.05).On the 1st,4th,and 7th days of injury,group A had obvious inhibitory effect on the increase of NO in tissues.Compared with groups B and C,there was no significant difference in inhibiting the increase of NO content(P>0.05).(4)SOD activity in injured tissue: intra-group comparison:The changes of SOD activity in the modeling groups of the four groups showed a gradually increasing trend,and there were significant differences between the three drug groups on the4 th day and the 7th day(P<0.05).Comparison between groups: Compared with group D,the SOD activity in the three groups was significantly higher on the 4th and 7th day,with significant difference(P<0.05).The drugs in group A could significantly enhance the activity of SOD,and there was no significant difference compared with groups B and C(P>0.05).(5)MDA content in injured tissue: compared within the group,the MDA content in the injured tissue of each model group showed a gradual downward trend.Compared with the E group,the MDA content was higher on the 1st and 4th day,with statistical significance(P <0.05).Comparison between groups,compared with group D,the content of MDA in the three groups was significantly decreased,and the difference was significant(P<0.05).There was no statistical significance between the two(P>0.05).Conclusion: The intervention of Jiawei No.2 Huoluo ointment can significantly improve the symptoms and signs of acute soft tissue injury in rats.Jiawei No.2 Huoluo ointment,Erhuang Xinshang Zhitong ointment and Voltaren ointment can treat acute soft tissue injury in rats with comparable efficacy.The mechanism of action of Jiawei No.2 Huoluo ointment in the treatment of acute soft tissue injury in rats may be related to inhibiting the increase of NO concentration in local tissue,enhancing SOD activity and reducing tissue MDA content,which can enhance the antioxidant activity of local tissue in rats,reduce damage and promote repair. |
