| Research purposes: To establish an acute soft tissue injury model in SD rats,and to observe the effect of Jiawei No.2 Huoluo ointment on the injury symptom index,histomorphological changes and the contents of TNF-α,IL-1β and IL-1β in damaged tissues in rats.Influence changes,and compared with Erhuang Xinshang Zhitong ointment and the commonly used clinical drug Voltaren,to explore the therapeutic mechanism of Jiawei No.2 Huoluo ointment on acute soft tissue injury,to provide a certain basis for the clinical application of the drug,and to strengthen the treatment of Zheng’s injury.Excavation and research of prescriptions.Research methods: 96 male SD rats were selected in this experiment,and after 1week of adaptive feeding,the acute closed soft tissue injury model was made by heavy blow method.The SD rats after successful modeling were randomly divided into five groups(n=18),namely group A(Jiawei No.2 Huoluo ointment group),group B(erhuangxinshangzhitong ointment group),group C(Voltarin group),group D(model control group),group E(blank group).Regularly and quantitatively externally applied drugs,and collected materials in batches on the 1st,4th,and 7th days after successful modeling,and observed and recorded the injury syndrome index,changes in muscle tissue morphology,TNF-α,IL-1β,PGE-2 content,all data were processed and analyzed by SPSS25.0.Research results: In terms of injury symptom index: each modeling group had significant differences compared with the blank group(p<0.05),indicating that the modeling was successful;there were significant differences between the drug groups and the D group on the 1st,4th,and 7th days(p<0.05).0.05),indicating that each drug group can improve the swelling,ecchymosis,dysfunction and other symptoms of injured limbs,and there is no significant difference in the scores between the drug groups on the 1st,4th,and 7th days(p>0.05).,In terms of histomorphology: Compared with the model group,each drug group can significantly reduce the inflammatory response of the injured local soft tissue and promote the repair of muscle fibers.In terms of the content of TNF-α in the tissue,the trend of each modeling group increased first and then decreased,compared with the group E,there were significant differences(p <0.05).The content of TNF-α was significantly higher than that of each drug group;group A could significantly inhibit the increase of TNF-α on the 1st,4th and 7th days of injury,and compared with groups B and C,there was no significant difference in inhibiting the increase of TNF-α.difference(p>0.05).In terms of IL-1β content in tissues: on the first day,each modeling group showed an upward trend first,reached a peak on the 4th day,and showed a downward trend on the 7th day.Compared with the E group,there were significant differences(p < 0.05).Comparison between the two groups: the content of IL-1β in the damaged tissue of each drug group was lower than that of group D,and there was a significant difference(p<0.05);each drug group could significantly inhibit the increase of IL-1β on the 1st,4th,and 7th days of injury.There was no significant difference in inhibiting the increase of IL-1β between the drug groups(p>0.05).In terms of the content of PGE-2 in the tissue,compared within the group,the trend of each model group on the 1st,4th,and 7th days showed a trend of first increasing and then decreasing.Compared with the E group,the values all increased and there were significant differences(p<0.05).Comparison between groups:The content of PGE-2 in each drug group was significantly higher than that in group D on the 1st,4th,and 7th days.There was no significant difference in inhibiting the increase of PGE-2 among the three drug groups(p >0.05).Conclusion: Jiawei No.2 Huoluo ointment can significantly improve the swelling,ecchymosis and other symptoms of acute soft tissue injury,inhibit the inflammatory response,and speed up the recovery of injured tissue.The curative effect is comparable,and its therapeutic mechanism may be related to the inhibition of the production of TNF-α,IL-1β and PGE-2 in the tissue. |
