Mongolian medicine Garidi-13 Pill,also known as Zhachong shisanwei pills,is composed of Terminalia chebula Retz.Radic aconiti kusnezoffll Preparata,Acorus tatarinowii,Radix aucklandiae,Moschus,Corallium rubrum,Margaritifera,Syzygium aromaticum Merr.Myristica fragrans Houtt.Aquilaria sinensis Gilg,Red claystone,Magnetitum,Glycyrrhiza uralensis Fisch.The main functions are expelling wind and dredging orifices,relaxing muscles and activating blood circulation,calming and removing "Xie Riwu Su".It is applied in treatment of hemiplegia,unclear speech,muscle and bone pain,rheumatism and joint pain.However,the chemical composition and mechanism of Zha Chong shisanwei pills are not clear.Based on the innovative theory of quality markers of traditional Chinese medicine,this study find and track the quality markers of Zhachong shisanwei pills from the chemical group,body composition,taste attribution and network pharmacology,clarifies the material basis of Zhachong shisanwei pills,and provides the research basis for improving the quality standard control of Mongolian medicine.In this study,chemical constituents of Zhachong shisanwei pills were systematically identified using ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)and gas chromatography-mass spectrometry(GC-MS).Then the blood and tissue samples after administration were analyzed.Then the electronic nose and electronic tongue technology were employed to study the medicinal properties of each single herb in Zhachong shisanwei pills.Finally,the quality markers of Zhachong shisanwei pills were selected through the network pharmacology in the main components.This study was divided into the following five parts:1 Identification of chemical constituents of Zhachong shisanwei pills.In this study,UPLC-Q-TOF-MS/MS and Peakview/Masterview data processing software were used to identified the chemical constituents of Zhachong shisanwei pillss.Mass spectrometry was collected in positive and negative ion mode respectively.A total of 98 compounds were identified,of which 17 were from Terminalia chebula Retz.,mainly tannins and phenolic acids,20 from Glycyrrhiza uralensis Fisch,mainly flavonoids,15 from Myristica fragrans Houtt.mainly flavonoids and volatile oils,10 from Corallium rubrum,,mainly flavonoids,12 from Syzygium aromaticum Merr.,mainly volatile oils,7 from Aquilaria sinensis Gilg,,mainly 2-(2-phenylethyl)chromones,7 from Radix aucklandiae,,mainly Sesquiterpenes,5 from Margaritifera,mainly amino acids,8 from Acorus tatarinowii,mainly volatile oils.3 from Radic aconiti kusnezoffll Preparata,mainly alkaloids,and3 from musk,mainly volatile oils.The volatile oil components of Zhachong shisanwei pills were analyzed by GC-MS,and the total ion flow diagram of the n-hexane extract was obtained and retrieval on computer mass spectrometry data system(NIST11 database),the mainstream components were deducted,and the compounds with matching value more than 70 were selected by area normalization method.A total of 41 components were identified,accounting for 57.11% of the total peak area.The main ingredients are eugenol,muscone,β-asarone,acetyleugenol.2 Analysis of blood components of Zhachong shisanwei pills.On the basis of identifying the chemical components of Zhachong shisanwei pills,the blood ingredient were studied.SD rats were gavaged with extract of Zhachong shisanwei pills and the plasma samples of SD rats were collected.The chromatographic column Shim-pack GIST C18(4.6 × 150 mm,5 um)was employed,methanol(mobile phase A)and 0.05% acetic acid water(mobile phase B)were used as mobile phase for gradient elution,and detected in(ESI)positive and negative ion mode of electrospray ion source.Then the chromatographic data of Zhachong13 pill extract,blank plasma and plasma after administration were compared by Peakview/Masterview software.31 prototype compounds,including Agarotetrol,Costunolide,muscone,isoalantolactone,germacrone,a-asarone,chlorogenic acid.3 Studies of the tissue distribution of Zhachong shisanwei pills.On the basis of clarifying the chemical composition,blood ingredient of Z hachong shisanwei pills,the chemical components in the tissue of Zhachong sh isanwei pills were further studied.6 tissue samples including brain,heart,lung,spleen,kidney,liver were collected and detected under the positive and negati ve ion mode of electrospray ion source(ESI).The chromatogram data of Zhac hong shisanwei pills extract,blank tissue sample and tissue sample after admin istration were compared and analyzed by Peakview/Masterview software.The re sults showed that 11 components were detected in brain,10 in heart tissue,12 in lung tissue,10 in spleen tissue,13 in kidney tissue,11 in liver tissue.Th ere are many distributions of agarotetrol,costunolide,liquiritigenin,germacrone, a-asarone,Chlorogenic acid,aconitine,glutamate.4 Study of the medicinal properties of Zhachong shisanwei pills--study on the characterization of single herb in Zhachong shisanwei pillss based on electronic tongue and electronic nose.Heracles Ⅱ electronic nose and Astree electronic tongue technology were employed to characterize the single herb of Zhachong shisanwei pills,and the data were analyzed by Alpha soft software.37 kinds of samples,including 10 medicinal materials extract,23 monomer composition solution and 4 control sample solution,were selected in the experiment.The principal component analysis was employed to distinguish and cluster the gas and taste of different samples.The results showed that each single herb(Myristica fragrans Houtt.Radic aconiti kusnezoffll Preparata,Radix aucklandiae,Moschus,Acorus tatarinowii,Aquilaria sinensis Gilg,Corallium rubrum,Glycyrrhiza uralensis Fisch,Terminalia chebula Retz.Syzygium aromaticum Merr.extract)did not interfere with each other.According to the results of principal component analysis,verify the firm ran ten single herbs can be divided three pills,basically made clear the smell of the monomer composition,An electronic nose and an electronic tongue to identify the odour components dehydrodiisoeugenol,aconitine,Benzoylaconitine,Costunolide,isoalantolactone,a-asarone Eugenol,germacrone.5 The mechanism of Zhachong 13 Pill in the treatment of stroke by using network pharmacologyUsing STRING and GENECARD database search firm at Zhachong shisanwei pills of main components targets and cerebrovascular disease related targets,the compound-disease both common targets as a key target,establish a drug target protein-disease target protein interaction(PPI)networks,build production "into the blood components-targets-pathway" network diagram,and will be the key targets for import David database KEGG pathway analysis,the results showed that the main targets of Zha Chong Shisanwei pill in the treatment of cerebrovascular diseases include ALB,CASP3,MMP-2,MMP-3,MMP-9,PLG,SELP,ESR1,ESR2,AR,IGF-1R,KDR,HSPA1 A,JAK2,MAPK,etc.Mainly related to inflammation and immune of the signaling pathways,hormone related pathways,complement and blood coagulation cascade pathway,VEGF signals article 25 related signaling pathway.Integrated the "chemical group-body composition-sexual flavour of belongin g-network pharmacology" various research results,found that 19 components of Costunolide,isoalantolactone,germacrone,liquiritigenin,liquiritin,ellagic acid,quercetin,chlorogenic acid,a-asarone,dehydrodiisoeugenol,myrislignan,muscon e,hyperoside,paeonol,aconitine,benzoylmesaconine,benzoylhypacoitine,agarot etrol,argininemay be the quality markers of Zhachong shisanwei pills,It lays a foundation for further expanding the research on quality control and pharmac odynamic substance of Zhachong Shisanwei Pills.
