Bergamot is the dried fruit of Citrus medica L.var.sarcodactylis Swingle,a medicinal and edible homologous plant of the Rutaceae family.It has the effects of soothing the liver and regulating qi,harmonizing the stomach to relieve pain,and eliminating dampness and phlegm.It is a commonly used medicine for soothing the liver and regulating qi in clinical Chinese medicine.It is often used to treat chronic gastritis,depression and other diseases.This study intends to comprehensively grasp the quality of the economic crop bergamot in the northeast of Chongqing by analyzing the chemical components of bergamot in the northeast of Chongqing,screening the chemical components that play a major role in chronic gastritis using the network pharmacological method,and conducting quantitative analysis of the relevant components in bergamot in different planting bases in the northeast of Chongqing.1.The chemical components of bergamot in the northeast of Chongqing were analyzed by UPLC-Q-Orbitrap HRMS.Based on the literature reports,32 chemical components were identified as the material basis of bergamot for the treatment of chronic gastritis,including 12 coumarins,14 flavonoids,3 organic acids,and 3 limonin;2.Based on network pharmacology,32 chemical component related targets were predicted using databases such as Swiss Target Prediction.Targets associated with chronic gastritis were collected through databases such as Gene Cards and OMIM,and a target protein interaction network was constructed using the STRING database;GO functional analysis and KEGG pathway enrichment analysis were carried out through DAVID database to explore the mechanism of bergamot in treating chronic gastritis.According to the analysis of the degree of chemical components,the chemical components that play a key role in the treatment of chronic gastritis were obtained,namely quercetin,luteolin,kaempferol,isorhamnetin,etc.3.Establish a high-performance liquid chromatography method for the simultaneous determination of the key active ingredients of bergamot in the treatment of chronic gastritis:quercetin,luteolin,kaempferol,and isorhamnetin.Conduct methodological validation on linearity,precision,stability,repeatability,and sample recovery.Through repeatability experiments,it was found that kaempferol was not detected in the sample.Considering the influence of pre-treatment methods,low content,and other factors,kaempferol was not included in this content determination experiment,Use quercetin,luteolin,and isorhamnetin as the target components for content determination.The contents of quercetin,luteolin and isorhamnetin in bergamot samples from Wanzhou,Yunyang,Liangping and Shizhu in the northeast of Chongqing were simultaneously detected,and the content of hesperidin was determined according to the content determination method under bergamot in Volume I of the Chinese Pharmacopoeia(2020 Edition).The quality analysis was carried out from the contents of quercetin,luteolin,isorhamnetin and hesperidin in each batch of samples.The results show that the content of each component in high-quality samples is high,but there are also some samples with low content of hesperidin and high content of other components,or with high content of hesperidin and low content of other components.The content correlation analysis of the four components showed that only quercetin and isorhamnetin were significantly positively correlated,and the other components were not significantly correlated,indicating that the content determination experiment was of practical significance and could make up for the inadequacy of the single component hesperidin to evaluate the quality of bergamot.In summary,this paper selects bergamot from the northeastern region of Chongqing as the research object.The material basis of bergamot is studied using ultra-high performance liquid chromatography four stage rod electrostatic field orbital trap high-resolution mass spectrometry technology.Network pharmacology research methods are used to screen chemical components that play a key role in the clinical treatment of bergamot-chronic gastritis,and their content is tested to comprehensively evaluate the quality of bergamot in the northeastern region of Chongqing.The research results provide reference for the quality standards of bergamot and have guiding significance for further development and utilization of bergamot.
