Lamiophlomis rotata(Benth.)Kudo,also known as dubutong,it is one of the precious medicinal plants in Qinghai Tibet Plateau.The study found that it has many pharmacological activities,such as bacteriostasis,anticancer and inhibiting tumor proliferation.However,the pharmacological mechanism of Lamiophlomis rotata(Benth.)Kudo is not clear and there is a great controversy.In this study,taking the Tibetan medicine Lamiophlomis rotata(Benth.)Kudo as the research object,the chemical components of Lamiophlomis rotata(Benth.)Kudo were analyzed by liquid chromatography-mass spectrometry,and the metabolic differences of flavonoids,iridoid glycosides and phenylethanol glycosides of Lamiophlomis rotata(Benth.)Kudo in rats were studied.The internal mechanism of Lamiophlomis rotata(Benth.)Kudo was systematically and deeply explored.Combined with animal models,the anti-inflammatory effect of Lamiophlomis rotata(Benth.)Kudo on mice with ulcerative colitis was studied,and its anti-inflammatory mechanism was preliminarily discussed,in order to provide support for the secondary development of Chinese and Tibetan medicine and provide scientific theoretical basis for clinical application.The main results of this study are as follows:1.Study on the chemical constituents of Lamiophlomis rotata(Benth.)KudoUltra-high performance liquid chromatography tandem time-of-flight mass spectrometry(UPLC-Q-TOF-MS/MS)method was isolated to identify the main chemical components,and their structures were identified according to the reference compound comparison mass spectrometry,retention time or temporarily assigned empirical molecular formula in the literature and database.Seventy-five of these chemical components were identified,mainly flavonoids,phenylproins,cycloether terpenoids and other types of compounds.2.Study on metabolism of Lamiophlomis rotata(Benth.)Kudo extract in ratsUPLC-Q-TOF-MS/MS technology was used to study the differences of metabolites of main active components in Lamiophlomis rotata(Benth.)Kudo extract in rats,so as to promote the disclosure of pharmacodynamic material basis and action mechanism of Chinese and Tibetan medicine.The results showed that the phenylethanolic glycosides and flavonoids of Lamiophlomis rotata(Benth.)Kudo were mainly oxygenated metabolites,and the iridoid components were mainly deoxymetabolites.The phenylethanolic glycosides in Angelica sinensis have short blood entry time and rapid blood drug metabolism.Through the comparative analysis of the abundance of phenylethanol glycosides,the phenyethanoside components are mainly caffeinated acyl or aduyl formed after enzymatic hydrolysis,and then form a series of derivatives.Flavonoids enter the blood for a long time,and enter the blood in the form of flavone aglycones,which may be that flavone glycosides first form aglycones in gastrointestinal digestion and then enter the blood.Iridoid glycosides have few blood components and low blood drug concentration.The blood components are rapidly metabolized in a short time.The research shows that the metabolic site of iridoid glycosides mainly occurs in the gastrointestinal tract,and there are few components that play a role in blood drugs.3.Study on anti-ulcerative colitis effect and mechanism of Lamiophlomis rotata(Benth.)KudoDextran sulfate(DSS)was used to explore the general activity,body weight,colon length changes,biochemical indicators and inflammatory factors expression in dss-induced ulcerative colitis mice.The results showed that Lamiophlomis rotata(Benth.)Kudo extract could significantly improve the weight changes of mice induced by DSS,inhibit the shortening of colon and reduce the pathological score of colon.At the same time,the extract of Lamiophlomis rotata(Benth.)Kudo could significantly inhibit the m RNA expression of NF-κB in mouse colon tissue caused by DSS,and reduce the concentration of TNF-α,IL-1βand IL-6 in the plasma of mice with ulcerative colitis induced by DSS.It shows that Lamiophlomis rotata(Benth.)Kudo can effectively treat acute UC in mice by inhibiting the NF-κB signaling pathway and enhancing the intestinal mucosal barrier function.
