Objective:Objective to explore the relationship between polymorphisms of IFN-γ,IL-4 and IL-12 A introns and susceptibility of patients with recurrent aphthous ulcer and their relatives.Methods:October 2018 to October 2020,29 mi RAU patients and 54 of their first degree relatives were selected from the Department of Stomatology,the first hospital of Shanxi Medical University.Fasting peripheral blood was collected and DNA was extracted by polymerase chain reaction(PCR).The corresponding loci of IFN-γ(rs1861494),IL-4(rs2227284)and IL-12A(rs76830965)were amplified.The distribution of genotypes and alleles was obtained by two-way sequencing.Pearson chi square was used to test whether the distribution of genotypes was in accordance with Harwin equilibrium.The routine clinical data and the distribution of genotypes and alleles of the three loci were compared between RAU probands,its confirmed relatives and healthy relatives.Statistical analysis was used to analyze the correlation between the three SNPs and the susceptibility of patients with recurrent aphthous ulcer and their relatives.Results:1.By comparing the clinical data of patients,there was no statistical significance in gender,vegetable and fruit intake(P < 0.05);however,the age of RAU probands was generally younger than that of their relatives(P < 0.05);the content of folic acid and Zn in RAU confirmed relatives was lower than that of their healthy relatives(P < 0.05)The levels of vitamin B12 and vitamin B12 were significantly lower than those of healthy relatives(P < 0.05).2.There was no significant difference in genotype and allele of IFN-γ rs1861494(C>T)among the groups(P > 0.05).There were 38 cases of CC genotype,including 10 cases of RAU proband,9 cases of RAU confirmed relatives and 19 cases of non RAU relatives;there were 33 cases of CT genotype,including 15 cases of RAU proband,9cases of RAU confirmed relatives and 9 cases of non RAU relatives;there were 12 cases of TT genotype,3 cases of each type.3.IL-12 A There was no significant difference in rs76830965(C > T)genotype and allele among the groups(P > 0.05).There were 45 cases of CC genotype,including 15 RAU probands,11 RAU confirmed relatives and 19 healthy relatives;28 cases of CT genotype,including 10 RAU probands,9 RAU confirmed relatives and 9 healthy relatives;10 cases of TT genotype,including 4 RAU probands,2 RAU confirmed relatives and 4 healthy relatives.4.IL-4 There were significant differences in rs2227284(T > C)genotype and allele among groups(P < 0.05).There were 29 cases of TT genotype,including 8 cases of RAU proband,5 cases of RAU confirmed relatives and 16 cases of non RAU relatives;there were 37 cases of CT genotype,including 10 cases of RAU proband,13 cases of RAU confirmed relatives and 14 cases of non RAU relatives;there were 17 cases of CC genotype,including 11 cases of RAU proband and 4 cases of RAU confirmed relatives Two cases were non RAU relatives.5.The genotypes of RAU patients and all their relatives conform to hardy Weinberg equilibrium law(P > 0.05).6.Based on the statistical difference of il-4rs2227284(T > C)genotype and allele,20 RAU nuclear families with both parents were selected for transmission disequilibrium test,and no statistical difference was found(P > 0.05).Conclusion:The polymorphism of IL-4(rs2227284)locus is associated with the susceptibility of RAU patients in Taiyuan and surrounding areas of Shanxi Province,and the population with C allele at this locus will increase the risk of the disease,but this locus has not been proved to be hereditary in the core families with RAU family history;IFN-(rs1861494)and il-12a(rs76830965)were not found to be associated with the susceptibility of RAU in this experiment,and a large number of samples from different races and populations were still needed to verify.
