Objective:This paper aims to explore the effect of Danbei Yifei Granules on food intake,lung coefficient,smad7 and miRNA-21 in pulmonary fibrosis rats,further understand the mechanism of Danbei Yifei Granules,and provide a new idea for clinical treatment of idiopathic pulmonary fibrosis.Method:1.In this experiment,the model was established by repeated stimulation and inhalation of bleomycin.2.120 male SD rats were randomly divided into 6 groups,with 20 rats in each group:control group(Group F),model group(Group D),methylprednisolone group(Group J),Danbei Yifei Granules high-dose group(Group A),medium-dose group(Group B)and low-dose group(Group C).3.After the model was successfully replicated,the rats were given routine intragastric administration once a day.On the 14th and 28th days after intragastric administration,rats in each group were randomly selected and killed.Lung wet weight was recorded by electronic balance,pathological sections were observed under microscope,the content of smad7 in lung was detected by Western-blot method,and the expression of miRNA-21 in lung was measured by RT-PCR.Food intake of rats was recorded.4.The data was analyzed by SPSS 23.0 statistical software,P<0.05 indicates statistical significance,and P<0.01 indicates significant statistical significance.Results:1.the general situation:the rats in the control group are strudy in body,quick to respond,with shiny fur,normal appetite,and stable breathing;the rats in the model group show listlessness,slow response to external stimuli,less movement,shortness of breath,and dark red secretions appear in in noses and mouths,sparse,dry and dull fur,part of the hair loss is serious,less food intake and slow weight growth compared with the control group.With the progress of time,the symptoms such as the decrease of activity and food intake in each administration group have improved to varying degrees and the weight gradually increased.2.pathological:①HE staining:In control group,the structure of lung is clear,complete and orderly.Compared with the control group,the alveolar structure of the model group is damaged to varying degrees,the space is obviously widened and the structure part is disregulated or collapsed,including infiltration of inflammatory cells.Compared with the model group,the degree of alveolitis in each administration group is improved to different degrees.The structure of alveolar in the methylprednisolone group is improved,but there were still inflammatory infiltration and edema.Compared with the methylprednisolone group,the alveolar in the Danbei Yifei groups are improved to a greater extent.② Masson staining:The structure of lung in the control group is complete and clear,the bronchial wall collagen layer is small,and a bit amount of collagen fibers stained in light blue are distributed in the alveolar area;In the model group,a large number of blue collagen fibers are observed in the outer wall of bronchus and blood vessels and in the thickened alveolar septum,alveolar wall is thickened and alveolar structure is damaged.Compared with the model group,the degree of pulmonary fibrosis in each treatment group is lighter,and the collagen blue staining area of lung interstitium,bronchial wall,blood vessel wall and alveoli is significantly reduced.Among them,the proportion of collagen staining consolidation area in each dose group of Danbei Yifei Significantly reduced,the consolidation area and alveolar blue-stained collagen deposition decreased,and it is significantly better than the methylprednisolone group.3.smad7:Compared with the control group,the expression of smad7 in lung of rats in the model control group was down-regulated.Compared with the model group,the expression of smad7 in lung was increased in Danbei Yifei groups.The difference between methylprednisolone group and the middle and high-dose groups of Danbei Yifei Granules had significant effects.4.miRNA-21:Compared with the control group,the expression of miRNA-21 in lung of rats in the model control group was increased.Compared with model group,miRNA-21 overexpression was inhibited in Danbei Yifei groups and pulmonary fibrosis was alleviated.The difference between methylprednisolone group and the middle and high-dose groups of Danbei Yifei Granules had significant effects.5.food-intake:The food intake of rats with idiopathic pulmonary fibrosis decreased and recovered after treatment,but the food intake of methylprednisolone group was significantly lower than that middle and high-dose groups of Danbei Yifei Granules.6.lung coefficient:The lung coefficient of Danbei Yifei groups were lower than that of methylprednisolone group and model group.Conclusion:1.Danbei Yifei Granules can better improve the mental state,restore the appetite of rats,reduce the degree of alveolitis and fibrosis and lung coefficient of idiopathic pulmonary fibrosis rats.2.Danbei Yifei Granules can inhibit the expression of miRNA-21.
