Objective: The study is aimed to study the characteristic of the bleomycin-induced pulmonary fibrosis in rats. And it is compared with the TCM pathogenesis of pulmonary fibrosis in clinic. So as to find the lab-proof of TCM pathogenesis of pulmonary fibrosis. To study the effect of Feixian Prescription(FXP) treating bleomycin-induced pulmonary fibrosis in rats, and connect with the pathogenesis of the model to study how FXP works in the disease. FXP is summarized from the years of clnical practice and theory study by my advisor Jiang Liangduo. It is composed with the hypha and powder of Dongchongxiacao, raw Huangqi, Hongjingtian, Mahuang, Yinxing, Gualou, Shuizhi, and Haigeqiao. FXP was set down to fix the mechanism of the disease and it has the effects of invigorating lung and kidney, eliminating phlegm and promoting blood circulation to remove meridian obstruction.Method: To copy the fibrosis model by infusing bleomycin (1ml/kg) through tracheal intubation. Choose prednisone as the positive drug. 100 Wistar rats were randomly divided into operation group with 85 infused bleomycin (1ml/kg) through tracheal intubation and fake operation group with 15 rats infused normal saline of the same volum. All the operation rats were randomly divided into model control group with 29, prednisone control group 28, and FXP group 28. Each were randomly divided into three time points. Observe the general and microcosmic change to study the connection between the changes and the pathogenesis of TCM to pulmonary fibrosis. First observe the spirit, appetite, activity, fur, weight, death, and any clinical situation that is possible. Second, measure the lung quotiety, observe the lung pathology with light microscope, measure the content of hydroxyproline in lung, to study the effect of FXP treating the bleomycin-induced pulmonary fibrosis in rats. Third, measure the transforming growth factor-β1 and platelet derived growth factor in blood serum to study the abc mechanism of FXP.Result: From the daily observation, find that the general state of FXP group is better than other model control group and prednisone control group. In the 45 days experiment no death case in FXP group and fake operation group, 4 death case in model control group, and 2 in prednisone control group, however, the statistics shows P>0.05, consider that it is related to the small sample and the experiment was not design only for the mortality rate, so we could not know weather the rats in former experiment will finish the whole 45 days observation. The lung quotiety is reduced quicklier in the FXP group, it descends markedly(P<0.05) in 28 time point compared to the 14 time point, while others descend markedly in 45 time point compared to the 14 time point. The lung pathology shows that the degree of fibrosis in FXP group is less than prednisone and model control group in 45 time point. Statistics shows FXP group's classification of fibrosis in the 45 time point is less than model group(P<0.05). the analysis of collagen with Masson staining also shows the surface density and digital density of collagen are less than that in model group and prednisone group... |
