Purposes:The death toll of lung cancer has been ranked first among malignant tumors for ten consecutive years,and lung adenocarcinoma has surpassed squamous cell carcinoma as the most common tissue type in lung cancer in recent years.Vascular invasion is one of the reasons for the high mortality rate of cancer.The aim of this research is to analyze liver kinase B1(LKB1),zinc finger E-box binding homeobox 1(ZEB1)expression and vasculogenic mimicry(VM)in lung adenocarcinoma and their relationships with clinicopathological parameters.In addition,the aim of this study is also to investigate the possible mechanism of LKB1 in the formation of VM in lung adenocarcinoma.Methods:LKB1,ZEB1 immunohistochemical marker staining,CD34/PAS double staining and AB/PAS staining were performed on the collected 161 lung adenocarcinoma tissue specimens and the corresponding adjacent tissues to observe the VM structure and the expression of LKB1 and ZEB1,and use statistical methods to analyze their differences between clinicopathological factors and prognosis.Results:The presence of VM and the positive rate of ZEB1 in lung adenocarcinoma tissue were significantly higher than the corresponding adjacent tissues,while the positive expression level of LKB1 in lung adenocarcinoma was significantly lower than that in the control group.Tumor WHO histopathological classification,the largest tumor diameter,lymph node metastasis,visceral pleura invasion and TNM stage are related with these factors.The expression of LKB1 is related to whether the tumor has mucin production.The postoperative overall survival of the VM structure,ZEB1 positive and LKB1 negative group was significantly less than that of the no VM structure,ZEB1 negative and LKB1 positive group.In the multivariate analysis,the largest tumor diameter,lymph node metastasis,visceral pleura invasion,WHO histopathological classification,and TNM stage are independent prognostic factors for the overall survival of patients with lung adenocarcinoma.In addition,the high expression of ZEB1 was positively correlated with VM,the expression of LKB1 was negatively correlated with VM,and the expression of LKB1 was negatively correlated with ZEB1.Conclusions:(1)The presence of VM,loss of LKB1 expression,and overexpression of ZEB1 promoted the growth and metastasis of lung adenocarcinoma.(2)Loss of LKB1 expression and overexpression of ZEB1 promoted VM formation in lung adenocarcinoma.(3)Patients with micropapillary,solid lung adenocarcinoma,large diameter,lymph node metastasis,high stage,visceral pleura invasion,LKB1 negative,ZEB1 positive,and VM had a poor prognosis.
