Objective To investigate the expression level and their clinical significance of LKB1mRNA and its gene product LKB1 in primary lung adenocarcinoma.Methods Using reverse transcription-polymerase chain reaction(RT-PCR) and SP immunohistochemistry, expression of LKB1mRNA and LKB1 was investigated in 48 primary lung adenocarcinoma tissue samples. Statistical analysis was done to analyze the relationship between the clinical data and the experimental results.Results 1. Immunohistochemistry: ?The positive expression rate of LKB1 in primary lung adenocarcinoma tissues (75.0%) was markedly lower than that in normal lung tissues(100%)( P<0.01).â‘¡The positive expression rate of LKB1 in well, moderately and poorly differentiated lung adenocarcinoma were respectively 90.5 %, 73.3%, 50.0%,which were significantly different.â‘¢The positive rate of LKB1 in stage I~ II was observably higher than that in stage III ~ IV.â‘£But no correlations were found between the positive expression rate of LKB1 in cases with lymph nodes metastasis(68.2 % ) and that in thoses without lymph nodes metastasis(80.8%)( P>0.05).2.Reverse transcription-polymerase chain reaction(RT-PCR) : â‘ The positive expression of LKBlmRNA in primary lung adenocarcinoma tissues was 72.9%, which was markedly lower than that of LKB1mRNA in normal lung tissues(97.9%). . â‘¡The positive expression of LKBlmRNA in well, moderately and poorly differentiated lung adenocarcinoma were respectively 90.5%, 73.3%, 41.7%, which were significantly different(P<0.05).â‘¢The positive rate of LKBlmRNA in stage...
