Objective:In this study,the effect of Weiweikang,a traditional Chinese medicine based on nourishing qi,nourishing yin and promoting blood circulation,on the Wnt/β-catenin signal pathway in rats with chronic atrophic gastritis(CAG)was observed to reveal its mechanism of treating chronic atrophic gastritis.To provide new ideas and new methods for the treatment of chronic atrophic gastritis by traditional Chinese medicine in the future.Methods:Twenty of 123 healthy male rats were randomly selected as the normal group,and the rest were used as models.Rats with chronic atrophic gastritis were reproduced by gavage with 20%sodium salicylate and combined with hunger and satiety factors.After successful modeling,they were randomly divided into 5 groups:Chinese medicine Weiwei Kanggao(3g/kg/d),medium(1.5g/kg/d),low(0.75g/kg/d)dose group and vitamin enzymes.Group(0.3g/kg/d),the normal group was administrated with 2ml/d saline.Observe the pathological changes of gastric mucosa in each group with HE staining and detect the protein expression of TNF-α,Wnt3,Wnt3α,Wnt5α,Wnt8α,β-Catenin and APC.Results:1.According to the experimental method,the hair of rats with chronic atrophic gastritis was dull during the modeling procession.After treatment,the rats’ weight increased significantly and their mental state improved.2.Criteria for successful CAG model replication:atrophy of gastric glands or inflammatory response,whitening of gastric mucosa,flattening or disappearance of gastric mucosal folds.HE staining was observed.After the modeling was completed,the gastric glands were atrophied or inflammatory.The gastric mucosa was whitened,and the gastric mucosa folds became flat or disappeared.HE staining observed that the gastric mucosal glands of the model rats atrophied,the number was reduced,the thickness of the lamina propria was reduced,and inflammatory cells were infiltrated.After treatment,compared with the chronic atrophic gastritis rat model group,the stomach of each treatment group rats mucosal atrophy improved significantly,the number of glands increased,the arrangement was tighter,the lamina propria thickened,and inflammatory cells decreased.3.IHC detected that compared with the normal group,the expression levels of TNF-α,β-catenin,Wnt3α,Wnt5α,Wnt3 and Wnt8αproteins in the model group increased(P<0.05),and the expression of APC was down-regulated(P<0.05),the difference is statistically significant.Compared with the model group,the expression levels of TNF-α,β-catenin,Wnt3α,Wnt5α,Wnt3 and Wnt8α were decreased in each treatment group(P<0.05),and the APC expression was increased(P<0.05).The difference was statistically significant.But Weiweikang high-dose group has the most obvious treatment effect,and it is better than retinoicin.Conclusion:Chinese medicine Weiweikang and retinomycin can improve chronic atrophic gastritis,but Chinese medicine Weiweikang is better than Weiweimycin in treating chronic atrophic gastritis,and the high-dose group has the best effect.The Chinese medicine Weiweikang may treat chronic atrophic gastritis by affecting the expression of WNT signaling pathway-related proteins and inhibiting the inflammatory response.
