Objective:To observe the pathology of chronic atrophic gastritis(CAG)rats with Weiweikang at different treatment stages and concentrations by observation and H-E(hematoxylin-eosin staining);Using IHC(immunohistochemical)to observe the changes of the related proteins Glil,Gli2,Gli3,Shh,Smo,SuFu in Hedgehog(Hh)signaling pathway and CyclinDl,in order to explore the therapeutic mechanism of Weiweikang on treating chronic atrophic gastritis.Methods:The rat model of CAG was established by the sodium salicylate and the irregular diet.After the model was made successfully,they were divided into five groups:model group、Weiweikang high(3 g/kg/d)、medium(1.5 g/kg/d)、low dose group(0.75 g/kg/d)and Victoria Su-enzyme group(0.3 g/kg/d).Then the rats were given corresponding concentration of drug treatment,and the model group and the normal group were given saline by gavage.The pathological changes of gastric mucosa in CAG rats were observed by H-E,and the changes of Glil,Gli2,Gli3,Shh,Smo,SuFu and CyclinDl were detected by IHC before and after treatment.The IOD value was analyzed by ipwin32 and the experim ental datas were processed statistically by SPSS17.0.Results:①Before treatment,some glands of gastric mucosa atrophied or disappeared,some of them had intestinal metaplasia,and some had inflammatory cell infiltration.After treatment,the number of gastric glands increased by H-E,the arrangement was neater,the symptoms of intestinal metaplasia improved,and inflammatory cells decreased;②Compared with model group,Glil,Gli2,Gli3,Shh,Smo,SuFu and CyclinDl in gastric mucosa of rats in high,medium and low dose groups of Weiweikang and Victoria Su-enzyme group were decreased(P<0.001);③Compared with Weiweikang medium dose、Weiweikang low dose group and Victoria Su-enzyme group,the protein expression of Glil,Gli2,Gli3,Shh,Smo,SuFu and CyclinDl in the Weiweikang high-dose group was decreased evidently(P<0.001).Weiweikang and Victoria Su-enzyme can threat CAG model group,but the Weiweikang high-dose group shows the best effect.Conclusion:The therapeutic effect of Weiweikang on chronic atrophic gastritis has been demonstrated and may be due to the influence related factors on Hedgehog signaling pathway. |
