Objective:Based on the liver main drainage theory,the effects of JWSNS and its dissolving formula group on stress resilience of chronic unpredictable mild stress rats were investigated,and the possible mechanism of action was explained.Methods:The sucrose preference test(SPT),open field test(OFT)and forced swimming test(FST)were used to observe the behavioral changes of rats with slow cums.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)was utilized to detect the expression levels of miR-124,REST mRNA and SCP1 mRNA in the hippocampus of rats.Content of REST and SCP1 protein in rat hippocampus was detected by Western blot.Immunofluorescence assay was used to detect the expression of Brdu-DCX and NeuN in rat hippocampus DG region.Results:1.In CUMS group,sucrose preference was significantly decreased(P<0.01),total traveling distance was significantly decreased(P<0.01),and immobility time was significantly prolonged(P<0.05).JWSNS could significantly improve sucrose preference(P<0.01),increase total traveling distance(P<0.01),and shorten immobility time(P<0.05).JWSNS can substantially increase the proportion of stress elasticity rats in the group.2.JWSNS could significantly increase the number of Brdu-DCX(P<0.01)and NeuN(P<0.01)labeled positive cells in hippocampus DG region of cums rats.3.JWSNS significantly increased miR-124 content(P<0.01),decreased REST mRNA content(P<0.05)and SCP1 mRNA content(P<0.05)in the hippocampus of cums rats.JWSNS could decrease REST protein content(P<0.05)and SCP1 protein content in hippocampus(P<0.05).4.Under the SPT standard,the ratio of stress resilient rats in the JWSNS group was 14/15,that in the QPF group was 11/13,and that in the SGF group was 10/13.Under the OFT standard,the ratio of stress resilient rats in the JWSNS group was 13/15,the ratio of stress resilient rats in the QPF group was 8/13,and the ratio of stress resilient rats in the SGF group was 10/13.Under the SPT standard,the ratio of stress resilient rats in the JWSNS group was 14/15,the ratio of stress resilient rats in the QPF group was 9/13,and the ratio of stress resilient rats in the SGF group was 9/13.5.Compared with cums group,sucrose preference in QPF group and SGF group was significantly increased(P<0.01,P<0.01).Compared with JWSNS group,there was not any statistical difference in sucrose preference between QPF group and SGF group.Compared with cums,the total traveling distance of open field in QPF group and SGF group was significantly increased(P<0.05,P<0.01).Compared with JWSNSS group,the total distance of open field in QPF group and SGF group had no statistical difference.Compared with the CUMS group,the immobility time of rats in the QPF group and the SGF group was significantly reduced(P<0.05,P<0.01).Compared with the JWSNS group,the immobility time of rats in the QPF group and the SGF group was not statistically different.Conclusions:1.In the process of cums,there are differences between stress resilient rats and stress susceptible rats;JWSNS could significantly increase the proportion of stress resilience in rats and improve behavioral abnormalities in cums rats.2.JWSNS can increase the number of Brdu-DCX and Neun labeled positive cells in hippocampus DG region,protect hippocampus neurons and promote hippocampus neurogenesis.3.JWSNS can significantly increase the level of miR-124 in the hippocampus of cums rats,thus improving the stress resilience of the body.4.The effect of JWSNS on enhancing the body stress resilience was significantly better than it’s SGF and QPF;in the process of chronic stress,the pathological state of insufficient liver drainage and excessive liver drainage coexist.5.JWSNS promote hippocampus neurogenesis by regulating miR-124 content through REST-SCP1 loop,which may be one of the mechanisms by which JWSNS improve the body’s stress resilience. |
