| Objective:1.Objective to study the differences of Bifidobacterium,Lactobacillus,Bacteroides and Escherichia coli in feces of patients with polycystic ovarian syndrome(PCOS)with phlegm dampness type,PCOS patients without phlegm dampness type and healthy women,and to analyze the correlation between the contents of the four kinds of intestinal bacteria and PCOS;2.Objective to investigate the clinical efficacy of electroacupuncture intervention in the treatment of PCOS of phlegm dampness type and its influence on the content of the four kinds of intestinal bacteria.Methods:Study 1: 20 cases of PCOS with phlegm dampness type,20 cases of PCOS without phlegm dampness type and 20 cases of normal control group were selected respectively.The general data of the patients were collected,the basic endocrine blood hormone levels were measured,transabdominal or transvaginal color Doppler ultrasound was performed,and the number of bilateral ovarian basal sinus follicles was recorded.Fecal samples were collected,and the contents of Bacteroides,Escherichia coli(intermediate bacteria)as well as Bifidobacteria,Lactobacillus(beneficial bacteria)in fecal samples were detected by real-time fluorescence quantitative PCR technology(SYBR Green I dye method)to study the differences of intestinal flora among the three groups.Study 2: 64 patients with PCOS with phlegm dampness type were selected and randomly divided into acupuncture and medicine group and traditional Chinese medicine group with 32 cases in each group.The traditional Chinese medicine group took Cangfu Daotan Decoction 3-5 days after menstruation.The acupuncture and medicine group was treated with Cangfu Daotan Decoction combined with electroacupuncture.At the same time,after 3 months of treatment,the changes of BMI,endocrine level,the number of bilateral ovarian basal sinus follicles and the improvement of clinical symptoms were observed,and the ovulation rate of the two groups were compared.The feces of all the patients in the morning were collected.The contents of Bacteroides,Bifidobacteria,Lactobacillus and Escherichia coli in fecal samples were detected by real-time fluorescence quantitative PCR(SYBR Green I dye method).The differences of intestinal flora between the two groups before and after treatment were observed.Results:1.The BMI of PCOS group with phlegm dampness type was significantly higher than that of PCOS group without phlegm dampness type and normal control group(P<0.05),but there was no significant difference between PCOS group without phlegm dampness type and normal control group(P>0.05).The levels of basal LH,basal LH / basal FSH,basal T and the number of bilateral ovarian basal sinus follicles in PCOS group of Phlegm dampness type and non phlegm dampness type were higher than those in normal control group(P<0.05),but there was no significant difference between PCOS group of phlegm dampness type and non phlegm dampness type(P>0.05).2.The contents of Bacteroides and Escherichia coli in phlegm damp PCOS group were higher than those in non phlegm damp PCOS group and normal control group(P<0.05),the contents of Bacteroides and Escherichia coli in non phlegm damp PCOS group were higher than those in normal control group(P<0.05).The content of Bifidobacterium and Lactobacillus in phlegm damp PCOS group was lower than that in non phlegm damp PCOS group and normal control group(P<0.05),the content of Bifidobacterium and Lactobacillus in non phlegm damp PCOS group was lower than that in normal control group(P<0.05).3.After treatment,BMI,basal LH,basal LH/basal FSH and the number of the bilateral ovarian basal sinus follicles in both the acupuncture and medicine group as well as traditional Chinese medicine group were lower than those before treatment(P<0.05),The difference between two groups was statistically significant(P<0.05).The basal T level in the acupuncture and medicine group was lower than that before treatment(P<0.05),there was no significant difference in the basal T level before and after treatment in the traditional Chinese medicine group(P>0.05).4.After treatment,the contents of Bacteroides and Escherichia coli in feces of the acupuncture and medicine group and the traditional Chinese medicine group were lower than those before treatment(P<0.05),the difference between two groups was statistically significant(P<0.05).The contents of Bifidobacterium and Lactobacillus were higher than those before treatment(P<0.05),and the difference between the two groups was statistically significant(P<0.05).5.After treatment,the TCM syndrome scores of both the acupuncture and medicine group and the traditional Chinese medicine group were lower than those before treatment(P<0.05),and the difference between two groups was statistically significant(P<0.05).The total effective rate of the acupuncture and medicine group was64.52%,and that of the traditional Chinese medicine group was 33.33%,and the difference between two groups was statistically significant(P<0.05).After treatment,the ovulation rate was 86.21% in the acupuncture and medicine group and 68.60% in the traditional Chinese medicine group,with statistically significant difference(P<0.01).Conclusions:1.The contents of Bacteroides and Escherichia coli(intermediate bacteria)in the feces of PCOS patients with phlegm dampness type increased significantly,while the contents of Bifidobacterium and Lactobacillus(beneficial bacteria)decreased significantly.2.Electroacupuncture intervention can significantly improve the clinical symptoms of PCOS patients with phlegm dampness type,reduce BMI,regulate basal LH and T levels,improve ovulation rate,and effectively strengthen the clinical efficacy of traditional Chinese medicine in the treatment of PCOS patients with phlegm dampness type.3.Electroacupuncture intervention can significantly increase the regulatory effect of traditional Chinese medicine on the contents of Bifidobacterium,Lactobacillus,Bacteroides and Escherichia coli in the intestinal flora of patients with PCOS of phlegm dampness type. |
