To Study The Effect Of Cang Fu Dao Tan Pill On PCOS Of Phlegm Dampness Type And The Influence Of Intestinal Flora

Objective:In this study,we observed the clinical effect of cangfu dao tan pill on polycystic ovary syndrome（PCOS）,evaluated its effect objectively,and analyzed the characteristics and changes of intestinal flora of PCOS before and after treatment with cangfu dao tan pill by using 16S rDNA sequencing technology.Methods:According to the principle of random control,90 cases of PCOS with phlegm dampness type were divided into three groups:Chinese medicine group,western medicine group and Chinese and Western medicine group.The traditional Chinese medicine group was treated with cangfu dao tan pill,the western medicine group was treated with daying-35orally,and the Chinese and Western medicine group was treated with daying-35 combined with cangfu dao tan pill.After 3 months of treatment,the TCM syndrome effect,waist hip ratio,body mass index（BMI）,Estrodiol（E₂）,luteinizing hormone（LH）,follicle stimulating hormone（FSH）,LH/FSH,anti Mullerian hormone（AMH）were compared among the three groups and testosterone（T）were used to compare the comprehensive clinical efficacy of the three groups of patients.At the same time,fecal samples of the patients in the Chinese medicine group before and after treatment were collectedin the non menstrual period.16S rDNA sequencing technology was used to analyze and describe the characteristics of intestinal flora of the patients before and after treatment.Results:（1）Before treatment,there was no significant difference in age,BMI,waist hip ratio,menstruation,AMH,E₂,LH,FSH,LH/FSH,T and TCM syndrome score among the three groups（P>0.05）.（2）After treatment,the symptoms of the three groups were improved,and the TCM syndrome score was reduced compared with that before treatment.The total effective rate of the three groups was 72.41%,55.17%and 96.55%respectively（P<0.05）.（3）The total effective rate of the three groups was 75.86%in the traditional Chinese medicine group,82.75%in the western medicine group and 93.10%in the traditional Chinese and western medicine group（P<0.05）.（4）The intestinal microflora of patients with phlegm dampness PCOS were significantly different in microbial species and microbial abundance after treatment.（5）Cangfu dao tan pill has a certain effect on the intestinal flora of patients,mainly affecting the metabolism and abundance of Gram-negative bacteria and potential pathogenic bacteria in intestinal microorganisms.（6）There was a correlation between the relative abundance of intestinal microflora and the clinical parameters BMI,AMH,T value and LH/FSH ratio of phlegm dampnessPCOS.Conclusion:（1）The results show that cang fu dao tan pill has clinical effect in the treatment of phlegm dampness PCOS,and its combination with western medicine Diane-35 has definite clinical effect in the treatment of phlegm dampness PCOS,and there is no adverse reaction in the treatment process;（2）the results show that cangfu dao tan pill has influence on the types of intestinal flora and the abundance of different microorganisms in phlegm dampness PCOS;（3）The results of this study provide a microbial theory for the treatment of phlegm dampness PCOS.
