| Objective:The effects of Jinshui Liujun Decoction and its disassembled prescriptions on COPD model rats with Yin deficiency and phlegm syndrome were studied.The changes of serum IFN-gamma,IL-lbeta,IL-4 and TNF-alpha levels were detected.The changes of mucin MUC5AC and MUC5B gene expression were observed.The mechanism of Jinshui Liujun Decoction in treating COPD Yin deficiency and phlegm syndrome was discussed.The experimental basis for clinical application of Jinshui Liujun Decoction in treating COPD Yin deficiency。Methods:Ten of 60 male SD rats of clean grade were randomly selected as blank group and the other 50 as model group.The COPD rat model of yin deficiency and phlegm syndrome was established by intratracheal instillation of lipopolysaccharide+smoking+intramuscular injection of dexamethasone.After successful modeling,rats in the model group were randomly divided into model group,Jinshui Liujun Decoction group(whole prescription group),Yin-nourishing group,phlegm-resolving group and azithromycin group,with 10 rats in each group.In the blank group,rats in the model group were given saline intragastric administration,and the other four groups were given the same amount of corresponding drugs for 14 consecutive days.The general situation of rats in each group was observed dynamically during the whole experiment.Rats were executed 12 hours after the last gastric perfusion and blood and lung tissue samples were collected.The inflammation and fibrosis of lung tissue were observed by hematoxylin-eosin(HE)staining and Mason staining;the number of goblet cells was calculated by PAS staining;the expression of MUC5AC and MUC5B protein in lung tissue was observed by immunohistochemistry;the changes of IFN-gamma,IL-lbeta,IL-4 and TNF-alpha in serum were detected by ELISA;and the gene table of mucin lung tissue(MUC5AC,MUC5B)was observed by RT-PCR.Results:1.Changes in general conditions of rats in each group:Compared with model group,the respiratory rate of rats in each treatment group was slowed down,asthma,irritability and other symptoms were alleviated,activity was increased,and stool condition was improved.2.Histopathological changes of lung tissue of rats in each group:There was no significant change in blank group.The model group showed obvious inflammatory cell infiltration.Compared with the model group,the inflammation and fibrosis of each treatment group were improved.3.Compared with the normal group,the content of IFN-gamma in the model group increased significantly(P<0.05),while the content of IL-lbeta,IL-4 and TNF-a had no significant difference.Compared with the model group,the IFN-gamma content in the whole prescription group,the Yin-nourishing group and the phlegm-resolving group decreased significantly(P<0.05);the TNF-alpha content in the whole prescription group and the Yin-nourishing group increased(P<0.05);the expression of IL-beta and IL-4 in each group had no significant difference.4.The number of goblet cells in bronchial epithelium in model group was significantly higher than that in blank group[(90.17+16.47)vs(16.00+10.20),P<0.05].The number of goblet cells in bronchial epithelium in whole prescription group,phlegm-resolving group and azithromycin group was lower than that in model group[(58.17+22.99)vs(90.17+16.47),(58.33+23.48)vs(90.17+16.47),(60.07+18.47).The number of goblet cells in bronchial epithelium of rats in Yin-nourishing group was not significantly different from that in model group[(87.07(+15.47)vs(90.17(+16.47).P>0.05];the number of goblet cells in Sputum-resolving group and azithromycin group had no significant difference[(58.17(+22.99)vs(60.07(+18.47)].(58.33(+23.48)vs(60.07(+18.47).P>0.05];and the number of goblet cells in whole prescription group,phlegm-resolving group and,all>0.05].5.The expression of MUC5AC and MUC5B:Compared with the blank group,the expression of MUC5AC and MUC5B in the model group increased significantly[(5.44+2.84)vs(1.08+0.74),(5.61+1.91)vs(1.09+0.56),P<0.05].Compared with the model group,the expression of MUC5AC and MUC5B in the whole prescription group,phlegm-resolving group,Yin-nourishing group and azithromycin group were all down-regulated(P<0.05).Compared with azithromycin group,the expression of MUC5AC and MUC5B in the whole prescription group was significantly decreased(P<0.05).CONCLUSION:It is suggested that there are inflammatory reactions and high expression of MUC5AC and MUC5B genes in COPD rats with Yin-deficiency and phlegm-decoction syndrome.Jinshui Liujun Decoction can reduce IFN-gamma content,mucin MUC5AC and MUC5B secretion,and inhibit the expression of MUC5AC and MUC5B in COPD Yin-deficiency phlegm... |
