Objective:The study was designed to investigate the effect and miR-34a-5p/ERK/pERK involved mechanism of acupuncture on NK cell mediated cytotoxicity(NKCC)of SD rat model of chronic fatigue syndrome(CFS).Methods:1.The effect of acupuncture on NKCC of CFS rat model:30 male rats were divided randomly into normal group,model group and acupuncture group.Model of Chronic fatigue syndrome was induced by combined methods of exhaustive swimming and chronic constriction for 21 days.Acupuncture were applied to acupuncture group at “ ganshu”(BL18),“pishu”(BL20),“ shenshu”(BL23)and “zusanli ”(ST34)for 15min/d and lasted for 3 weeks while normal group and model group were strained for the same time.NKCC was detected by LDH releasing method。2.The miR-34a-5p/ERK/pERK involved mechanism of acupuncture on NKCC of CFS rat model:(1)ERK1/2,pERK1/2 and miR-34a-5p in NK cells of the same 30 rats mentioned above were detected by Western blotting and qRT-PCR method respectively.(2)After model establishing,20 male rats were divided randomly into antagomir group and saline group.miR-34a-5p antagomir or saline of the same volume were injected intraperitoneally to antagomir group or saline group,followed by the same treatment of acupuncture group.NKCC,ERK1/2,pERK1/2 and miR-34a-5p in NK cells were measured.Results:1.Compared to normal group,NKCC of model group was significantly reduced(P=0.014),acupuncture group showed strongly increased NKCC compared to model group(P=0.050),no significant difference found between normal group and acupuncture group.2.(1)Compared to normal group,miR-34a-5p 、ERK1/2 and pERK1/2 of model group decreased significantly(P=0.008,P<0.001,P=0.017).Compared to model group,acupuncture group showed significantly elevated ERK1/2 、pERK1/2 and miR-34a-5p(P<0.001,P=0.032,P=0.044).(2)Antagomir group showed significantly reduced NKCC and miR-34a-5p than saline group(P=0.005,P=0.025),ERK1/2 and pERK1/2 decreased,but not statistically significant(P=0.680,P=0.052).Conclusion:1.NKCC decreased significantly in CFS rats,but increased strongly after acupuncture.2.ERK1/2 、 pERK1/2 and miR-34a-5p in CFS rat NK cells reduced significantly,but were improved by acupuncture strongly.Acupuncture may increase NKCC by improving miR-34a-5p/ERK/pERK-independent pathway,thus regulating abnormal immune state in CFS.
