Part Ⅰ The correlation between mitochondrial function of CGCs and ovarian reserveObjective: To investigate the correlation of mitochondrial function of cumulus granulosa cells around human oocytes(CGCs)with ovarian reserve.Methods: A total of 94 women underwent in vitro fertilization/ intracytoplasmic sperm injection and embryo transfer(IVF/ ICSI-ET)at the reproductive medicine center of 901 hospital of PLA from October 2018 to December 2019 were enrolled this study.According to the ovarian reserve,the subjects were divided into two groups : normal ovarian reserve group(62 cases)and diminished ovarian reserve group(32 cases),CGCs of a single MⅡoocyte in the patients of each group were collected on the day of oocyte retrieval.Mitochondrial transcription factor A(TFAM)m RNA expression level and mt DNA content were measured using Real-time Quantitative Polymerase Chain Reaction(Q-PCR),mt DNA4977 bp loss rate were detected using ordinary PCR,and ATP content were measured using luciferase method.All indexes above and the outcomes of IVF were compared between DOR and NOR groups.Results: The content of mt DNA and ATP,TFAM m RNA expression level in the DOR group were significantly lower than that in the NOR group,and the mt DNA4977 bp loss was not detected in both groups.There was no significantly difference in the high quality embryo rate between the two groups.Conclusion: Mitochondrial function of CGCs is correlated with ovarian reserve.Part Ⅱ The correlation between mitochondrial function of CGCs and oocyte maturityObjective: To investigate the correlation of mitochondrial function of cumulus granulosa cells around human oocytes(CGCs)with oocyte maturity.Methods: CGCs from 303 oocytes of the women underwent ICSI at the reproductive medicine center of 901 hospital of PLA from October 2018 to December 2019 were collected separately.According to oocyte maturity,they were divided into three groups: MⅡgroup(186),MⅠgroup(78)and GV group(39).Mitochondrial transcription factor A(TFAM)m RNA expression level and mt DNA content were measured using Real-time Quantitative Polymerase Chain Reaction(Q-PCR),mt DNA4977 bp loss rate were detected using ordinary PCR,and ATP content were measured using luciferase method.All indexes above were compared among three groups.Results: The expression level of TFAM m RNA,content of mt DNA and ATP increase progressively with the oocyte maturity,there was significantly difference in the content of ATP among the three groups,and the mt DNA content in GV group was significantly lower than that in MⅡ group.The mt DNA4977 bp loss was not detected in three groups.Conclusion: The contents of mt DNA and ATP were correlated with oocyte maturity.
