Objective:To study effects of different doses of Semiaquilegia granule(SG)on serum sex hormones,vascular endothelial growth factor(VEGF),integrin-β3(ITG-β3)and rat embryo implantation in rats with bad endometrial receptivity of kidney-deficiency blood-stasis type(Abbreviation Model Rat).To explore the effect of Semiaquilegia granule on endometrial receptivity and its possible mechanism.Methods:96 SPF-level mature female SD rats were divided randomly into blank group(A group),model group(B group),low-dose SG group(C1group),medium-dose SG group(C2 group)and high-dose SG group(C3 group)and progynova group(D group),16 cases in each group.Rats in each group except the A group were given hydroxyurea(450mg/kg/d)intragastrically and epinephrine(0.3mg/kg/d)subcutaneously injected with to establish a rat model of endometrial intolerance due to kidney deficiency and blood stasis.At the same time of modeling,the C1 group,C2 group,and C3 group were administered with 3.125 g/kg,6.25 g/kg,and 12.5g/kg SG.The D group was administered with 0.104 mg/kg/d progynova,while A group and B group were administered with same amount of normal saline.After the modeling was completed,female and male rats were placed in cages at a ratio of 2:1,and penile or vaginal smear examinations were performed the next day.Those who found sperm under the microscope were recorded as the first day of pregnancy,while those without sperm were kept in cages until sperm were found.On the5th of pregnancy,8 rats were randomly selected from each group.After anesthesia,abdominal aortic blood was collected,and uterus was striped.The levels of serum estrogen(E2)and progesterone(PROG)were detected by Enzyme-linked immunosorbent assay(Enzyme-linked immunosorbent,ELISA).HE staining was applied to observe morphological changes of endometrium tissue and vascular distribution.The immunohistochemistry was applied to detect expression of VEGF and ITG-β3 in endometrial tissue.On the 8th of pregnancy,8 rats were selected from each group.After anesthesia and uterus was striped,the number of pregnant rats and number of implantation points were calculated.Results:Compared with A group,on the 5th day of pregnancy,The expression levels of serum E2 and PROG in the B group rats were reduced,the endometrial lesions were significantly reduced,and the number of blood vessels was significantly reduced.The levels of VEGF and ITG-β3 in the rat endometrial tissue were significantly reduced(P<0.05).Compared with B group,after treatment with D group and C2 group,C3 group endometrial lesions were improved significantly,number of blood vessels,levels of serum E2 and PROG,expression levels of VEGF and ITG-β3 in endometrium tissue were increased significantly(P<0.05).Compared with A group,on the 8th day of pregnancy,although there was no statistical difference in the pregnancy rate of rats in each group(P>0.05),the average number of implanted blastocysts in the B group was significantly reduced,and the difference was statistically significant(P<0.05).Compared with B group,the average number of implantation blastocysts of C1,C2,C3 group and D group was significantly increased(P<0.05).Conclusion:1.Semiaquilegia granule can increase the expression of estrogen and progesterone in model rats and improve the morphology of endometrium in model rats;2.Semiaquilegia granule can improve the expression level of VEGF in the endometrium of model rats,increase the number of endometrial blood vessels,and improve the endometrial blood supply;3.The difference between Semiaquilegia granule and Estradiol valerate was not statistically significant. |
