ObjectiveThis paper aims to investigate the effects of grain-moxibustion and estrogen(E2)in the treatment of thin endometrium and their possible mechanisms.MethodsForty healthy SPF mature female SD rats were randomly divided into four groups:normal group,model group,estrogen group and grain-moxibustion group.The normal group was not modeled,and the model group,the estrogen group and the grain-moxibustion group are all established thin endometrial model during estrus.The normal group was fed normally,and the model group was given 2 ml of normal saline the second day after modeling,once a day;The estrogen group was given 2 ml of estradiol the second day after modeling,once a day.The grain-moxibustion group was given the "Guanyuan" on the abdomen and the "Shenyu" on both sides of the back of the second day after modeling.Every acupuncture point was given 7 strong,once a day The endometrial tissue was taken after three consecutive intervention estrus cycles and the endometrial tissue morphology was observed by HE staining.The thickness of endometrial tissue was measured.Immunohistochemistry was used to detect the expression of keratin,vimentin and vascular endothelial growth factor(VEGF)in endometrial tissues.Western blot was used to detect the expression levels of LIF and HOXA10 proteins in endometrial tissues.ResultsThe endometrial thickness of the model group was significantly thinner than that of the normal group(P<0.01).Compared with the model group,the endometrial thickness of the estrogen group and the wheat moxibustion group increased significantly(P<0.05).The average thickness of endometrium in the grain-moxibustion group was higher than that in the estrogen group,but the difference was not statistically significant(P>0.05).The expression of endometrial keratin in the model group was significantly lower than that in the normal group(P<0.01).Compared with the model group,the expression of endometrial keratin in the estrogen group and the moxibustion group was significantly increased(P<0.01).The expression of vimentin in the endometrium of the model group was significantly lower than that in the normal group(P<0.01).Compared with the model group,the expression of vimentin in the endometrium of the estrogen group and the moxibustion group was significantly increased.(P<0.01).The expression of VEGF in the endometrium of the model group was significantly lower than that in the normal group(P<0.01).Compared with the model group,the expression of VEGF in the endometrium of the estrogen group and the grain-moxibustion group was significantly increased(P<0.01).The expression levels of keratin,vimentin and VEGF in the grain-moxibustion group were higher than those in the estrogen group,but the difference was not statistically significant(P>0.05).The expression levels of HOXA10 and LIF protein in the endometrium of the model group were significantly lower than those in the normal group(P<0.01).Compared with the model group,the expression of HOXA10 and LIF protein in the endometrium of the estrogen group and the grain-moxibustion group were significant increase in levels(P<0.05).ConclusionGrain-moxibustion can improve endometrial thickness,promote endometrial growth and improve endometrial receptivity-related factors such as HOXA10 and LIF by up-regulating the expression of keratin,vimentin and VEGF in rat endometrium.The level of improvement in endometrial receptivity plays a role in repairment. |
