| Objective:The increase in uterine cavity manipulation and other factors has led to an increase in endometrial damage,and thin endometrium has become more common.However,the pathogenesis of thin endometrium is still unclear.There is a lot of evidence that angiogenesis is closely related to endometrial repair.Therefore,this study established a thin endometrial rat model,to observe the effect of wheat-grain moxibustion at "Guanyuan" and "Zigong" on angiogenesis related factors and Notch signaling pathway related molecules in rats with thin endometrium,and explore its possible mechanism for treating thin endometrium from the perspective of angiogenesis.Methods:Thirty SPF-grade healthy adult female SD rats were divided into 3 groups according to the random number table:control group,model group,and wheat-grain moxibustion group,with 10 rats in each group.Except for the control group,rats in the other two groups were injected with 95%ethanol into the uterus to establish a thin endometrial model during estrus.The control group and the model group were fixed daily after anesthesia,and the wheat-grain moxibustion group was given wheat-grain moxibustion at the acupoints of "Guanyuan"(CV4)and"Zigong"(EX-CA1)the next day after the model was established,7 moxa cones for each acupoints.After 3 estrus cycles,samples were taken during the estrus period of rats,and HE staining was used to observe the changes in endometrial thickness and uterine morphology,the expression of vascular endothelial growth factor(VEGF)and angiopoietin-2(Ang-2)were detected by immunohistochemistry and real-time fluorescent quantitative PCR(RT-qPCR),the protein and mRNA expression of Notch receptor(Notch1)and Notch ligand(DLL4)were detected by Western blot and RT-qPCR.Results:(1)Endometrial thickness and number of glands and blood vessels:compared with the control group,the endometrial thickness of rats in the model group was significantly thinner,the number of endometrial glands and blood vessels were significantly reduced(P<0.01);while compared with the model group,the thickness of endometrium in the wheat-grain moxibustion group was thicker,and the number of endometrial glands and blood vessels was increased significantly(P<0.05,P<0.01).(2)Expression of angiogenesis related regulatory factors VEGF and Ang-2:the MOD value of VEGF and Ang-2 in the endometrium of the model group were significantly lower than that of the control group(P<0.01);compared with the model group,the wheat-grain moxibustion group was significantly higher than that of the model group(P<0.01,P<0.05)The mRNA expression of VEGF and Ang-2 in the endometrium of rats in the model group were significantly lower than that in the control group(P<0.01);compared with the model group,the wheat-grain moxibustion group mRNA expression was higher than the model group(P<0.05).(3)Expression of Notch signaling pathway Notch1 and DLL4:the protein expression of Notch1 and DLL4 in the endometrium of the model group were significantly lower than that of the control group(P<0.01);compared with the model group,the wheat-grain moxibustion group increased significantly(P<0.05).The mRNA expression of Notchl and DLL4 in the endometrium of rats in the model group were significantly lower than those in the control group(P<0.01);compared with the model group,the wheat-grain moxibustion group was significantly increased(P<0.05,P<0.01).Conclusion:Wheat-grain moxibustion "Guanyuan" and "Zigong" acupoints can improve the morphology of the endometrial tissue of thin uterine model rats and repair the damaged endometrium;it is possible that the mechanism is to affect the expression of angiogenesis related factors VEGF and Ang-2 in the endometrium of rats with thin endometrium by up-regulating the expression of Notch signaling pathway related molecules in the endometrium,promote the angiogenesis of the endometrium,improve the thickness of the endometrium and achieve the repair effect. |
