Objective:To detect the differential expression of key inflammatory factors proteins and mRNAs in TLR4/NF-κB,and to explore the possible pathogenesis of TLR4/NF-κB regulation of Xinjiang Han and Uygur phlegm retention type syndrome,and to analyze the possible factors of the pathogenesis of hyperuricemia.Methods:General data of 139 patients with hyperuricemia in the Xinjiang Uygur Autonomous Region Chinese Medicine Hospital,Kashi First People’s Hospital outpatient department and inpatient department from November 2016 to August 1818 were collected.They were divided into Han group and Uygur group according to Chinese medicine.The diagnostic criteria for syndrome type are classified into phlegm retention type syndrome and non-phlegm retention type syndrome,complete the information collection form;collect blood samples and detect UA,GLU,TC,TG,HDL-C,LDL-C,BUN,CR content;The expression levels of MyD88,NF-kB,TNF-α,IL-1β protein and mRNA in the samples were detected by enzyme-linked immunosorbent assay(ELISA)and reverse transcription-polymerase chain reaction(RT-PCR).Results:1.The expression levels of MyD88,NF-κB,TNF-αand IL-1β in Han and phlegm retention type patients were significantly higherthan those in Uygur and non-phlegm retention type;2.Patients with phlegm retention type syndrome The expression levels of MyD88,NF-κB and TNF-αmRNA in Chinese Han population were significantly lower than those in Uygur;3.MyD88,NF-κB,TNF-α,IL-1β protein expression and body weight,BMI,CR,TC,TG,BUN The content was weakly correlated;the expression of MyD88,NF-κB,TNF-α,IL-1β mRNA was negatively correlated with TC,GLU and BUN,and positively correlated with LDL-C.Conclusion:It is speculated that "TLR4/NF-κB" may regulate the expression of inflammatory factors MyD88,NF-κB and downstream inflammatory factors TNF-α,IL-1βprotein and mRNA in the middle and upper reaches,thereby regulating Xinjiang Han and Uygur hyperuricemia.The onset of sputum turbidity blockphlegm retention type syndrome;inflammatory factors MyD88,NF-κB,TNF-α,IL-1β may be the influencing factors of hyperuricemia. |
