| In recent years,the incidence of cardiovascular and cerebrovascular diseases is increasing year after year,the formation of arteriovenous thrombosis is the basis of many cardiovascular disease,which is one of the main causes to lead people to death.Currently,anticoagulant drugs such as heparin and warfarin commonly used in clinical have still exist some limitations,which would cause some side-effect to bady.Therefore,it is necessary to study some new anticoagulant preparations from natural plants.Auricularia auricular is a famous edible fungi with medicinal value,and studies have shown that polysaccharide from Auricularia auricular has many activities such as anticoagulant,antitumor,hypolipidemic,hypolipidemic,hypoglycemic and so on.In this study,we used Auricularia auricular as raw material,extracted and screened polysaccharide with the best anti-coagulation activity.And selected five kinds of polysaccharides from natural plant to combine with polysaccharides that from Auricularia auricular to choose the strongest anticoagulant synergistic samples.Then,we took the experiment in vivo to anlaysis the safety and anticoagulant activity of it.The results as follows:(1)Extracted neutral,alkaline and acidic tree kind of polysaccharide from Auricularia aurisula by different solvents.The test of anticoagulant activity shows,acidic polysaccharide of Auricularia aurisula has the best anticoagulant activity.Compared the polysaccharide samples that prepared by ultrasonic,microwave,the results showed that M-ACAP had the best anticoagulant activity,and its sulfate content was the highest,the ability of inhibiting fibrinogen(FIB)to transform fibrin IC50 was 0.76±0.05 mg/mL,when the concentration of polysaccharide at 4.Omg/mL,the APTT was 125.03±1.64s,TT was 70.39±0.87s.(2)Determination the anticoagulant activity of Trp,TP,JP,GLP,SP.Used M-ACAP as based polysaccharides to combination with five kind of polysaccharides,the result showed M-ACAP:TP(1:1),M-ACAP:SP(1:1),M-ACAP:SP(1:2),M-ACAP:SP(1:4)had synergistic anticoagulant.From them,M-ACAP:SP(1:2)had the strongest synergistic anticoagulant,it’s combined index was 0.63175.When the concentration was 2.0mg/mL,the inhibition rate of fibrinogen(FIB)to fibrin was reduced to 91.06±0.79%,the APTT was 95.47±0.61s,TT was 62.24±0.53s.(3)CombiSPAP had no significant to body weight and organ index in vivo,but could increased the CT,BT,APTT and TT significantly.And it’s prolongation was correlated with polysaccharide concentration positively.These findings have provide the theoretical basis on the research and development of natural compound anticoagulant preparations. |
