Objective To study the extration optimization of polysaccharides from Auricularia auricular using uniform design and the effects of Auricularia auricular polysaccharides on scavenging oxygen free radical and anti-lipid peroxidation in vitro and the activity of SOD in serum.Methods Uniform Design was used to optimize the effects of processing parameters of extraction from Auricularia auricular on the yield of polysaccharides. Four independent variables such as extraction time (hour), solid:liquid ratio (g/ml), extraction times and extraction temperature (oC) were studied. The optimal conditions were determined from SPSS13.0. Superoxide anion free radical ((O2-)) induced by the auto-ox idation of pyrogallol and hydroxyl free radical (·OH ) induced by Fenton system were used to study the effect on scacenging oxygen free radical.The inhibition on lipid peroxidation malondialdehyde (MDA )of rat liver was determined by thiobarabituric acid (TBA ) colorimetric method. The rats were fed with polysaccharides from Auricularia auricular orally at the dose of 200mg/kg, then the contents of SOD in the serum of the rats were determined.Results The optimum conditions of polysaccharides extraction were liquid:solid ratio of 1:45, extraction time of 1 hour, water extraction temperature of 80oC, and water extraction times of 3 times. Under optimized conditions, the experimental yield4.3 % agreed closely with the predicted yield. Auricularia auricular polysaccharides could significantly scavenge (O2-)and·OH, and remarkably inhibit the lipid peroxidation of rat liver. The scavenging and inhibitory effect exhibited in a concentration-dependent manner in a specific range. It also significantly increased the contents of SOD in serum of rats after gastric perfusion.Conclusion The results indicated that possesses the effect of scavenging oxygen free radicals and anti-lipid peroxidation, which shows the protection of liver mitochondria.
