The Study Of ShenJing Guben Decoction About Antioxidation And Senile-resistant Function

Purpose:This study treats the antioxidation in vitro of Shen-Jing-Gu-Ben Decoction and the effect of the decoction acting on mice which are subacute aging models by injecting D-galactose.Then make sure the antioxidation of Shen-Jing-Gu-Ben Decoction and preliminarily elucidate the mechanism of antioxidation.This study provides experimental foundation for clinical application and exploiture as functional health food of Shen-Jing-Gu-Ben Decoction.Method:（1）60 mice were randomly divided into the normal control group,the model group,the low,middle,high dose group of Shen-Jing-Gu-Ben Decoction and Jin-Gui-Shen-Qi pill group.There were 6 groups in total,10 mice in each group.All mice were injected D-galactose（at the dose of 125mg/kg）at their subcutaneous tissue of nape,except the mice in normal control group.Comparing the treatment of Shen-Jing-Gu-Ben DecoctionShen-Jing-Gu-Ben Decoction with the treatment of Jin-Gui-Shen-Qi pill,we need to observe the ability of learning and memory in mice,to measure the percentage of liveweight growth,the index of thymus and spleen and to determine the content of SOD,MDA and T-AOC,etc,in brain tissue.（2）The study of antioxidation in vitro:we evaluated the clearance capability of free radical and antioxidation activity in vitro,through determining the clear rate of O₂ˉ,DPPH free radical and hydroxy radical.Result:（1）The time that the mice of Shen-Jing-Gu-Ben Decoction high,middle and low dose group used to find the goal was significantly increased reduced（P<0.01）.The percentage of liveweight growth of Shen-Jing-Gu-Ben Decoction high dose group was reduced（P<0.01）.The the index of thymus and spleen of Shen-Jing-Gu-Ben Decoction high dose group was significantly increased（P<0.01）.The spleen index of Shen-Jing-Gu-Ben Decoction middle dose group was slightly increased（P<0.05）.The activity of SOD and T-AOC of Shen-Jing-Gu-Ben Decoction high dose group was significantly increased（P<0.01）and the content of MDA was significantly reduced（P<0.01）.The activity of SOD of Shen-Jing-Gu-Ben Decoction middle dose group was slightly increased.（2）Shen-Jing-Gu-Ben Decoction has the clearance effect of DPPH free radical,O₂ˉand hydroxy radical in various degree.And this experimental result explained that Shen-Jing-Gu-Ben Decoction has some antioxidant effects.When its concentration was 1mg/ml,the clear rate of DPPH free radical,O₂ˉ,and hydroxy radical was 71.18%、66.58%、36.59%,by using Shen-Jing-Gu-Ben Decoction.Conclusion:Shen-Jing-Gu-Ben Decoction intervenced in senile mouse model and the result showed that the decoction had antioxidation and senile-resistant function..It can promote the antioxidant effects of brain tissue,to postpone the aging.The antioxidation in vitro of Shen-Jing-Gu-Ben Decoction is good.This study provides experimental foundation for clinical application and exploiture as functional health food of Shen-Jing-Gu-Ben Decoction.
