Objective:To study the effect of Guben Fangxiao Decoction(GBFXD) and its active ingredients, astragaloside, on α-SMA and INF-γ mRNA of IL-4 treated normal human bronchial epithelium cell.Methods:Cellular inflammation model was made by using IL-4 to stimulate normal human bronchial epithelium cell (BEAS-2B). The cells were divided into normal group, model group, the low-dose GBFXD group, the medium-dose GBFXD group, the high-dose GBFXD group, the low-dose astragaloside group, the medium-dose astragaloside group and the high-dose astragaloside group. The cells were collected separately after drug intervention till 24 hours. Real-time quantitative polymerase chain reaction (RT-PCR) was employed to confirm the expression of INF-γ at mRNA level and ELISA assay was employed to confirm the expression of α-SMA at protein level in cells. Finally data were analyzed and processed by using statistical methods.Results:1. The ELISA results showed that:the levels of protein expression changes in administration groups were lower than the model group, and they all showed obvious difference. 2. The PCR results shown that:compared with the normal group, the expression of INF-γ at mRNA level of the model group was significantly lower. Compared with the model group, all of the administration groups could up regulate the expression of INF-γ at mRNA level obviously.Conclusion:1. α-SMA was a special marker protein for airway remodeling and INF-γ was an important anti-inflammatory factor in the pathogenesis of asthma. They may regarded as one of laboratory parameters judge the disease changes; 2. Guben Fangxiao Decoction and its active ingredients, astragaloside, could down regulate the expression of α-SMA and INF-γ mRNA in IL-4 treated normal human bronchial epithelium cell:relieve airway remodeling and Inhibit the inflammatory response to relieve occurrence of asthma.
