Purpose Prostatic carcinoma(PCA)incidence has increasing in recent years,prostate specific antigen(PSA)and Gleason sum is mainly used to guide therapy and prognosis in PCA.PSA is released by prostate epithelial cell,PCA leads to the broken of blood-prostate barrier and cause PSA leak into the mesenchyme,PSA go through the capillary and lymphatic vessels and cause serum PSA rising[1,2,3].Utilize serum PSA to indicate the integrality of blood-prostate barrier.Analyze 29 prostatic carcinoma cases of relationship between claudin-4 and PSA determines,and discuss the relationship between the expression of claudin-4 in human prostatic carcinoma and blood-prostate barrier.Analyze the relationship between Gleason sum and claudin-4 expression,and analyze the relationship between Gleason sum and serum PSA determines,and discuss the relationship between the blood-prostate barrier and prostatic carcinoma differentiation.Methods 29 prostatic carcinoma cases were adopted from the First Affiliated Hospital of Chongqing Medical University with admission date during january,2008 to January,2015.The prostatic carcinoma tissue was utilized to detect claudin-4 with IH studies.Applying IPP software to analyze the expression of claudin-4 to obtain the IOD and AREA determines and figure out the claudin-4 expression determines by the formula of[IOD/(PPI-Area)].Applying radioimmunoassay to obtain the serum PSA determines before the surgery operation.Spearman rank correlation test was applied to analyzing the relationship between the expression of claudin-4 in prostatic carcinoma and PSA.Fishier exact probability was applied to analyzing the relationship of the expression of claudin-4 in prostatic carcinoma and serum PSA determines with Gleason score respectively.Adopting SPSS 11.0 software to analyze the data.Results The highest and lowest PSA determines is 152.5 μg/L and 0.98μg/L respectively among the 29 prostatic carcinoma cases.There are 7 and 15 prostatic carcinoma cases in PSA<10.0μg/L group and PSA>20.0μg/L group respectively.And the 1020.0μg/L group contains 7 prostate cancer cases.The highest and lowest Gleason sum is 9 and 5 respectively among the 29prostatic carcinoma cases.There are 4 and 15 prostatic carcinoma cases in Gleason sum 5 and 6 respectively.And the Gleason sum7 and 8 contains 6 and 3 prostatic cancer cases respectively.There is 1 prostatic carcinoma case in Gleason sum 9[4].The claudin-4expression determines less than 2.1 including 4 prostatic cancer cases.And the determines more than 2.25 contains 12 prostatic carcinoma cases while the determines between 2.1 and 2.25comprising 13 prostatic carcinoma cases.And the statistic analyzing indicated claudin-4 expression is significantly correlated to serum PSA in prostatic carcinoma(r=0.532,P=0.003).The statistic analyzing indicated the serum PSA is significantly correlated to Gleason score in prostatic carcinoma(P=0.027).Fishier exact probability analyzing results reveals that the expression of claudin-4 in prostatic carcinoma is connected to Gleason score and the difference is not statistically significance(r=0.2,P=0.8).Conclusion The claudin-4 expression is significantly correlated to serum PSA in prostatic carcinoma,PSA is corrected to blood-prostate barrier,Claudin-4 might contribute to blood-prostate barrier.And claudin-4 may be a maker of clinical diagnosis,classification,follow-up of prostatic carcinoma.The serum PSA is significantly correlated to Gleason score in prostatic carcinoma.With the Gleason sum rising,the greater the damage would did in blood-prostate barrier.There is no statistically significant evidence to show the expression of claudin-4 in prostatic carcinoma is relative to Gleason score. |
