Objective:To investigate the expression of claudin-2and claudin-10inbladder urothelial carcinoma and non-tumor bladder tissue,explore therelationship between their expression with bladder urothelial carcinomaoccurrence and metastasis,and the correlation between them.Methods: pathologically confirmed,50bladder urothelial carcinoma(25lymph node metastasis and25without lymph node metastasis)and50non-tumor bladder tissue were used to detect the expression of Claudin-2protein and Claudin-10protein by immuno-histochemical SP (StreptavidinPerdcidase)method, comparative analysis of the relationship between theirexpression levels and bladder urothelial cancer clinicopathologicalparameters.Results:1.The positive expression rate of Claudin-2in bladder urothelialcarcinoma and non-tumor bladder tissue were70%(35/50)、12%(6/50)respectively.the expression levels in the bladder cancer group wassignificantly higher than the non-tumor group (Ρ<0.05).The positiveexpression rate of Claudin-2in lymph node metastasis and without lymphnode metastasis were84%(21/25),56%(14/25) respectively.the expressionlevels in lymph node metastasis group significantly higher than thatwithout lymph node metastasis (Ρ<0.05).The expression levels of Claudin-2in different age、gender and tumor size groups were nosignificant difference (Ρ>0.05).There were significant difference indifferent clinical stage, histological grade group of bladder cancer(Ρ<0.05).2.The positive expression rate of Claudin-10in bladder urothelialcarcinoma and non-tumor bladder tissue were32%(16/50)、72%(36/50)respectively.the expression levels in the bladder cancer group wassignificantly lower than the non-tumor group (Ρ<0.05).The positiveexpression rate of Claudin-10in lymph node metastasis and without lymphnode metastasis were16%(4/25),44%(11/25) respectively.the expressionlevels in lymph node metastasis group significantly lower than that withoutlymph node metastasis(Ρ<0.05).The expression levels of Claudin-10indifferent age、gender and tumor size groups were no significant difference(Ρ>0.05).There were significant difference in different clinical stage,histological grade group of bladder cancer(Ρ <0.05).3.The expression levels of Claudin-2protein and Claudin-10protein inbladder urothelial carcinoma was negative correlation (r=0.050p=0.731).Conclusion:1.Claudin-2expression in the bladder cancer group wassignificantly higher than the non-tumor group,the expression levels inlymph node metastasis group significantly higher than that without lymphnode metastasis,prompted Claudin-2expression Up-regulation may promote the occurrence and metastasis of bladder urothelial carcinoma.2.Claudin-10expression in the bladder cancer group was significantlylower than the non-tumor group,the expression levels in lymph nodemetastasis group significantly lower than that without lymph nodemetastasis,prompted Claudin-10expression down-regulation or missingmay promote the occurrence and metastasis of bladder urothelialcarcinoma.3.The expression levels of Claudin-2protein and Claudin-10protein inbladder urothelial carcinoma was negative correlation.
