Purpose The serum adiponectin(APN)level in patients with gastric cancer(GC)group and control group were detected by protein microarray technology,we investigated decreased serum APN level affected clinical pathological characteristics of patients with GC,and furthermore investigated the correlations between APN level and clinical pathological characteristics of GC.Methods The modification of N-succinimidyl 4-(maleimidomethyl)cyclohexanecarboxylate(SMCC)protein microarray chip was optimized,and the characterization of SMCC protein chip was verified by Atomic Force Microscope(AFM)and Fourier Transform Infrared Spectrometer(ATR-FTIR).We tested serum APN level in 210 GC patients and 221 healthy cancer-free control subjects by SMCC modified protein microarray.Statistical analysis was performed using SPSS release 16 statistical package.Results The chemical functional groups modified by SMCC are stable in covalent bonding with gold surface,and self-assembled monolayer structure is successfully established.We found that the serum APN level was distinctly lower than that in the healthy control subjects(p<0.01).There was higher APN level in women than in men of the control subjects(p=0.024),but such a significant gender difference missed in the GC cases(p=0.265).Serum APN concentrations were higher in the older subjects(>55years)in both the controls(p=0.003)and the GC cases(p=0.014).We analyzed associations of serum APN levels and the clinical pathological characteristics in the GC patients and found that serum APN levels in the GC patients were distinctly correlated with tumor location(p=0.008),differentiation degree(p=0.011)and histologic type(p=0.006).No significant associations of serum ANP level with tumor size,invasion depth,nodal metastasis,distant metastasis,TNM stage,cancer embolus in vessels of gastric serosa were observed between the cases and the controls(p >0.05).Conclusion Our results revealed that serum APN might potentially reflect location,histogenesis and differentiation of GC.The biochip immunoassay would act as one substitute of conventional clinical assays for measuring serum APN in GC.
