Objective:To determine the contents of Bax protein,Bcl-2 protein,AChE,ChAT,SOD,MDA and GSH-Px in hippocampus of rats with dementia,gene expression of Bax and Bcl2,and behavioral expression in rats.To explore the effect of eight moxibustion methods and different moxibustion amounts on the above indexes,for eight moxibustion methods provide experimental evidence for the treatment of senile dementia.Methods:50 healthy SD rats were randomly divided into normal group,model group ai wick effect 3 zhuang moxa wick sticks 6 strong group the wick moxibustion 9 strong five groups,each group of 10 only in addition to the normal group,the rest of the group,all rats by intraperitoneal injection of9 AM every day for D-galactose 400 mg·kg-1,1 times a day,five weeks in a row,part cause subacute aging and dementia,starting from the 6 weeks for 3weeks according to the dosage of the 3 mg·kg-1 1 times daily intraperitoneal injection of scopolamine hydrobromide solution,the final preparation into AD model rats after the building using spirit turtle eight open hole moxibustion treatment 5 weeks after the end of day 1 start water maze rats behavior determination,five days before to platform to explore,the last 1 day to withdraw platform for space exploration water maze experiment over the next day to death group of rats,5 abdominal aortic venous blood serum,enzyme league immune method for determining the content of SOD,MDA,GSH-Px changes;The blood was collected and executed,the brain tissue was taken on the platform,the hippocampus was isolated,and AChE ChAT Bax bcl-2 was determined by enzyme linked immunoassay.The expression of Bax bcl-2gene was detected by rt-pcr.Results:⑴Behavioral expression of rats Compared with the normal group,the time spent in the model of dementia rats was significantly higher than that of the normal group,and the number of times of crossing the platform was significantly lower than that of the normal group,and the difference was statistically significant(p<0.01).Compared with the model group,three groups of treatment groups found that the platform time was reduced and the number of platforms increased through the platform,and the differences were statistically significant(p<0.01).⑵The hippocampal AChE ChAT Compared with normal group,the hippocampus AChE of rats was significantly increased,and ChAT was significantly decreased,with statistically significant differences(P<0.01).Compared with the model group,the hippocampal AChE in rats decreased significantly,and ChAT was significantly increased,and the differences were statistically significant(P<0.01).⑶Bax bcl-2 protein and gene expression in hippocampus of rats Compared with normal group,Bax protein and gene expression were significantly increased,and bcl-2 protein and gene expression were significantly decreased,and the differences were statistically significant(p<0.01).Compared with the model group,Bax protein content and gene expression were significantly decreased,and bcl-2 protein content and gene expression were significantly increased,and the differences were statistically significant(p<0.01).⑷Changes in serum SOD MDA GSH-Px in rats Compared with the normal group,the SOD in the serum decreased significantly,the MDA significantly increased,and the gsh-px decreased significantly,and the difference was statistically significant(P<0.01).Compared with the model group,the SOD in serum was significantly increased,MDA was significantly decreased,and gsh-px was significantly increased,and the difference was statistically significant(p<0.01).⑸The differences between the 3 groups were not significant(P>0.05),but the data of the 6 group and the 9 strong group were better.Conclusion:⑴Eight methods of Intelligent Turtle time point moxibustion can improve AD rat’s behavior expression,enhance memory,improve the symptoms of dementia to a certain extent.⑵Eight methods of Intelligent Turtle time point moxibustion can improve SOD and GSH-Px activity in AD rats,decrease the content of MDA.⑶Eight methods of intelligent turtle on acupuncture moxibustion can increase AD in rat hippocampus ChAT activity,reduce the activity of AchE.⑷Eight methods of intelligent turtle on acupuncture moxibustion can increase the expression of Bcl-2 protein and gene content in hippocampus of AD rats,reduce the content of Bax protein and gene expression⑸Different doses of the eight methods of Intelligent Turtle time point Moxa Moxibustion has little effect on AD rats,but increase the quantity of moxibustion can make better effect.
