This experiment was mainly to study the effects of different concentrations of cadmium(Cd)on the oxidative stress and lipid metabolism related genes in the Schizothorax prenanti,and to study the effect of Acidolysis oxidated konjac glucomannan(AOKGM).Experimental design: Added 0%(Cd treatment group)and 0.4% AOKGM(AOKGM group)to the diet of the Schizothorax prenanti,and after 8 weeks of feeding,then exposed the two groups of test fish to an aqueous environment with a Cd concentration of 0,100,250,and 1000 ?g/L for 15 days(temperature 0 to 10?).And the expressions of antioxidant,biochemical and lipid metabolism related genes in serum,hepatopancreas and mesonephros of each group were determined.The test results are as follows:1.Schizothorax prenanti had certain tolerance to different concentrations of Cd stress at low temperature(temperature 0~10 ?).Cd treatment group:With the increasing of Cd concentration,the activity of serum SOD,CAT and GSH showed a decreasing trend,and the activity of serum GSH-PX increased.Compared with Cd concentration of 0 ug/L,When Cd was 100~1000 ?g/L,it significantly increased the activity of SOD,CAT,GSH-PX in hepatopancreas,and SOD in mesonephros.When Cd was 250 and 1000 ?g/L,it significantly increased the activity of GSH in hepatopancreas,and GSH-PX in mesonephros.When Cd was 1000 ?g/L,the activity of CAT and GSH in the mesonephros was significantly increased.Cd caused oxidative stress,AOKGM had obvious intervention effect on oxidative stress,and there was no dose relationship with Cd.Cd treatment group:Different concentrations of Cd stress significantly increased MDA in the mesonephros and NO content in different tissues,and the oxidative stress of the mesonephros was more serious.AOKGM can reduce the content of NO and MDA to a certain extent.Intervention effects of AOKGM on changes of GSH and antioxidant enzymes in different tissues induced by cadmium were inconsistent.The related reasons needed to be further studied.2.Different concentrations of Cd caused the amino acid metabolism disorder of Schizothorax prenanti,which had an effect on hepatopancreas and mesonephros function.Cd treatment group : Compared with Cd concentration of 0 ug/L,when the Cd concentration was 100 ~ 1000 ?g/L,it significantly increased GPT activity in hepatopancreas,GOT activity in serum and mesonephros.When the Cd concentration was 250 and 1000 ?g/L,it significantly increased GOT activity in hepatopancreas,GPT activity and BUN content in serum.When the concentration of Cd was 100 and 1000 ?g/L,the activity of GPT in the mesonephros was significantly increased.AOKGM had a certain regulating effect on the amino acid metabolism caused by Cd.Compared with the Cd treatment group,GOT activity in serum and hepatopancreas of AOKGM group significantly decreased at different concentrations of Cd.When Cd was 250 ?g/L,the GPT activity of different tissues in the AOKGM group significantly decreased.When Cd was 1000 ?g/L,hepatopancreas GPT activity was significantly reduced in the AOKGM group.3.Different concentrations of Cd stress led to disorders of glucose metabolism.Cd treatment group:Compared with Cd concentration of 0 ug/L,when Cd was 100~1000 ?g/L,hepatopancreas and mesonephros LD content increased significantly,and serum Glu content increased significantly.When Cd was 250 and 1000 ?g/L,the Glu content in hepatopancreas significantly increased.When Cd was 100 ?g/L,the Glu content in the mesonephros significantly increased.AOKGM has a certain regulatory effect on glucose metabolism disorders.Compared with the Cd treatment group,When Cd was 250 and 1000 ?g/L,it significantly reduced serum Glu content,hepatopancreas Glu and LD content in AOKGM group.When Cd was 250 ?g/L,mesonephros Glu content were found significantly low in the AOKGM group.When Cd was 100 ?g/L,and execution with AOKGM group could significantly decrease Glu content in serum,mesonephros Glu and LD contents.4.Different concentrations of Cd were immunotoxic to Schizothorax prenanti.Cd treatment group:With the increasing of Cd concentration,serum ALP activity decreased before elevation,but hepatopancreas ALP activity increased,and serum LZM activity decreased.Compared with Cd concentration of 0 ?g/L,when Cd was 100 and1000 ?g/L,it increased ACP activity in serum and hepatopancreas.When Cd was 250 ?g/L,the activity of LZM in the mesonephros was significantly reduced.AOKGM can alleviate the immunotoxicity of Cd.The effect of AOKGM on the activity of ACP,ALP and LZM in different tissues is inconsistent,and the relevant mechanism needs to be further studied.5.Different concentrations of Cd caused disturbance of lipid metabolism in the Schizothorax prenanti.(1)The effects of Cd on TG and T-CHO indicators: In the Cd treatment group,compared with Cd 0 ?g/L,When Cd was 100~1000 ?g/L,serum T-CHO content was significantly decreased.Serum TG levels significantly elevated when Cd concentrations were 250 and 1000 ?g/L.When Cd was 1000 ?g/L,the contents of TG and T-CHO in hepatopancreas increased significantly.AOKGM can regulate the content of TG and T-CHO in varying degrees.(2)Effects of Cd on lipid metabolism related genes in hepatopancreas and mesonephros.Compared with Cd concentration of 0 ?g/L,when Cd was 100 ?g/L,it significantly up-regulated the relative expression of PPAR?,PPAR?,PPAR?,FAS,GPDH and HFABP in hepatopancreas and mesonephros,the relative expression of LPL and HSP70 in hepatopancreas,the relative expression of G6 PD in mesonephros.Exposed to 250 ?g/LCd up-regulated the relative expression level of PPAR??PPAR??PPAR??GPDH?HFABP and LPL in hepatopancreas and mesonephros,the relative expression level of HSP70?FAS in mesonephros.Exposed to 1000 ?g/LCd it caused significantly higher expression of HSP70?PPAR??PPAR??FAS?GPDH?HFABP and G6 PD,respectively,in the mesonephros.The results showed that different concentrations of Cd caused lipid metabolism disorder in Schizothorax prenanti.Compared with the Cd treatment group,when Cd was 100 ?g/L,the AOKGM group resulted in significantly decreased the relative expression of LPL in the hepatopancreas,the relative expression of G6 PD in the mesonephros.When Cd was 250 ?g/L,the AOKGM group can significantly induce the expressions of PPAR? in the hepatopancreas,the expressions of HSP70?FAS and LPL in the mesonephros.When Cd was 100 and 250 ?g/L,significant inhibition in the relative expression of HSP70?PPAR??PPAR??GPDH and HFABP was observed in the hepatopancreas at AOKGM group.When Cd was 100~1000 ?g/L,the relative expression levels of PPAR?,PPAR?,PPAR? and HFABP in the mesonephros of AOKGM group significantly down-regulated.AOKGM can slow down the toxicity of Cd and regulate lipid metabolism disorders.
