China, the only country in the world that owns larger-scale freshwater aquaculture which is very advanced than mariculture, has a vast water area. As the most developed country in freshwater fishery, China accounts for half of the global freshwater aquaculture consumption. With the increasing consumption, more and more questions arise, such as environment pollution caused by fisheries wastes irresponsibly disposed of and the low economic added value of freshwater fishery. To enhance the economic added value of fish-processing, a growing number of researchers start to exploit the comprehensive utilization of freshwater fishery resources, and it’s pretty helpful to the further development of freshwater aquaculture. Extracting collagen from fisheries wastes instead of the terrestrial animal skin e.g. cow and pig serves as a significant measure to augment the freshwater fish economic added value.Waters is wast, Schizothorax prenanti (Tchang) is a specialty of Ya’an Sichuan, commonly known as Ya fish. At present, there is a little studies about Schizothorax prenanti (Tchang) skin.In this paper, choose Schizothorax prenanti (Tchang) skin as the main raw materials in order to extract acid soluble collagen (ASC) and enzymatic skin collagen (PSC). at the same time, its part of identification were studied preliminary, which can provide some theoretical reference for Schizothorax prenanti (Tchang) skin collagen fully used and evelopment.Firstly, the extraction rate of Schizothorax prenanti (Tchang) fish skin collagen was regarded as a target to research the maximum extraction process conditions. Acid soluble collagen was extracted under the following conditions:the concentration of acetic acids was 0.56mol/L, liquid-to-solid ratio was 70:1, temperature was 41℃, the time of extraction was 50 h, so the maximum extraction rate of ASC was 942.707mg/g. Schizothorax prenanti (Tchang) fish skin collagen extraction factors from big to small was in turn for acetic acid concentration, temperature, liquid-to-solid ratio, time, which the acetic acid concentration and temperature Schizothorax prenanti (Tchang) fish skin collagen extraction effect is the most remarkable.Secondly, the extraction yield of Schizothorax prenanti (Tchang) fish slin collagen was regarded as a target to research the maximum extraction process conditions. PSC was extracted under the optimal conditions:liquid-to-solid ratio was 20:1, pH was 8,the amout of enzyme was 5000 U/g, the temperature was 55℃ reaction for 10 h, and the best extraction yield of ASC was 97.44%. In the view of extraction of collagen from Schizothorax prenanti (Tchang) skin, Schizothorax prenanti (Tchang) fish skin collagen extraction factors from big to small was in turn for temperature, liquid-to-solid ratio, pepsin dosage, pH, which the acetic acid concentration and temperature Schizothorax prenanti (Tchang) fish skin collagen extraction effect is the most remarkable.Finally, the identification of nature of ASC and PSC was studied, The maximum UV absorption wavelength of ASC and PSC of Schizothorax prenanti (Tchang) skin was 235 nm respectively. SDS-PAGE suggested that ASC of Schizothorax prenanti (Tchang) skin were comprised of al, a2 and P chain, which was characteristic of the triple helical conformation of the protein, but not PSC. And the molecular weigth of al was higher than a2. which might be classified as type I collagen. By amino acid analysis, ASC and PSC contain eight essential amino acids,which indicated that the basic amino acides have not been destroyed during the preparation of collagen, further confirmed the reasonableness of methed of preparation.
