Objective:To study the toxic effect of atmospheric PM2.5 on human umbilical vein endothelial cells and to explore its toxic effects on the cardiovascular system,and to provide reliable scientific basis for atmospheric pollution control and prevention of human cardiovascular diseases.Methods:The particulate matters(PM2.5)was collected in Shijiazhuang City;and eluted by the ultrasonic cleaning instrument;and desiccated through vacuum freeze-drying machine.The desiccated PM2.5 was prepared with PBS and DMEM medium into the following PM2.5 concentration:0,100,200 and400?g/ml.HUVEC-T1 cells were cultured in different PM2.5 concentrations respectively during a period of 6h,24h and 48h.The cell toxicity of PM2.5 was assessed by measuring the cell survival rate?MTT method?,the lactate dehydrogenase?LDH?and the cell proliferative ability?BrdU method?.The cell oxidative damage was assessed by measuring reactive oxygen species?ROS?,malondialdehyde?MDA?,superoxide dismutase?SOD?and catalase?CAT?.The cell inflammatory effect of PM2.5 was assessed by measuring extracellular IL-1?,IL-10,IL-12p70 and TNF-??CBA method?.Single factor analysis of variance was used to compare among groups,and Dunnett-t test was used to analyze the differences between the 0?g/ml group concentrations and other exposure groups,and the differences between the 24h,and 48h groups and the 6h group.Results:1.The comprehensive toxicity of PM2.5 on cells:Compared with0?g/ml group,the cell survival rate at 100,200,and 400?g/ml groups were decreased with the increasing of concentration,but the leakage of LDH was increased.Compared with 6h group,the cell survival rate at 24h and 48h groups were decreased with the increasing of the exposed time.But the leakage of LDH was increased.The cell proliferation capacity was consistent with the trend of cell survival rate,and decreased with the increasing of concentration and the exposure time.2.The oxidative damage effect of PM2.5 on cells:Compared with0?g/ml group,the production of ROS and MDA in cells at 100,200,and400?g/ml groups were increased with the increase of PM2.5.5 concentration but CAT and SOD activity was decreased with the increasing of concentration.Compared with 6h group,the production of ROS and MDA in cells at 24h and48h groups were increased with the exposure time,but CAT and SOD activity was decreased with the exposure time.3.The inflammatory effect of PM2.5 on cells:Compared with 0?g/ml group,the secretion of IL-1?,IL-10,IL-12p70 and TNF-?at 100,200,and400?g/ml groups were increased with the increasing of concentration.Compared with 6h group,the secretion of IL-1?at 48h group was increased significantly,while the secretion of IL-10,IL-12p70,and TNF-?at 24h and48h groups were no differences.Conclusions:PM2.5 can promote the oxidative stress response of HUVEC-T1 cells,and also induce the inflammatory responses,destroyed the ability of self-regulation,thus proceeded toxic effect on human umbilical vein endothelial cells,and a positive relationship of dose-response and time-effect was observed.
