Study And Application Of The Compound Chinese Medicine Extract On Androgenic Alopecia

In this paper,compound Chinese medicine extract was studied in vitro efficacy for multi target of androgenic alopecia(AGA)by using Chinese herbal medicine compound combined with the mechanism of androgenic alopecia occurred.The extract was prepared into the essence for anti-hair loss with the characteristics of refreshing,safe and effective.The patients with AGA can stick to it and achieve satisfactory result.Method for inhibiting 5α-reductase activity was established by UV Spectrophotometry.At 340 nm,Variation of the reaction solution was taken as the index after 10 min,and the inhibition rate was calculated according to the formula.In the test,5α-reductase reaction conditions were optimized to determine the optimum concentration of testosterone 20 μM,NADPH 40 μM,enzyme amount 300 μg.On this basis,the blank control group,positive control group,the experimental group were set.When the concentration of the extract was30 mg/mL,the inhibition rate was 63.35%,which was equivalent to that of finasteride 0.2mg/m L.The results showed the extract can inhibit 5α-reductase activity effectively.The MIC values of the extract were determined for S.aureus or other microorganisms that cause inflammation by continuous dilution method,and the value was measured by reciprocal of dilution multiple of the extract concentration.The results showed the range of MIC of the extract for S.aureus,P.acnes and M.furfur were 1/8~1/5,1/5~1/4,1/5~1/4 of the original extract concentration,and the corresponding drug concentration were 3.75~6.0mg/m L,6.0~7.5 mg/mL,6.0~7.5 mg/mL respectively.The datas showed the inhibitory effect was better than that of some other researchers.On the basis of the above experimental results,the extract was prepared into essence for anti-hair loss.The essence was tested by the GuangDong Foods and Cosmetics Testing Centre for health indicators and safety,that had reached the requirements of Hygienic Standard for Cosmetics and Safety of China.In the study of the essence promoted the mice skin microcirculation,forty Kunmingmices were randomly divided into four groups: high,medium and low dose of the extract were set as experimental group,and 15% ethanol was set as blank control group.The skin temperature of the mice before the experiment and at different time points(5,30,45,60,90min)after the experiment were measured by infrared thermometer,the results were based on difference of the temperature at different time points and before the experiment.The experiment found: the skin temperature of mice in each group was lower than that before the experiment,and there were significant difference after 5 min;After 30 min,the skin temperature of the blank control group was still lower than that before the experiment.The skin temperature of the mice in each experimental group was higher than that before the experiment,and there were significant difference;After 45 min,the skin temperature of blank control group was rising,high and middle dose groups were higher than that 30 min,the low dose group was decreasing,and there were significant difference;After 60 min,the skin temperature of the high dose group was still increasing,and the middle dose group showed a decreasing trend,all of which were significant difference;After 90 min,the skin temperature of the blank control group was still lower than the pre-experimental level,and the high dose group was also decreasing,but the high and middle dose groups were higher than the pre-experimental level,the low dose was basically restored the pre-experimental level,and there were significant difference.The results showed the essence can improve the skin temperature of mice and maintain the skin temperature rising time which was proportional to the dose of the essence,the finding indicate that compound Chinese medicine extract for anti-hair loss can improve skin temperature and promote skin microcirculation.C57BL/6J mices were used as experimental model to study the effects of essence on hair growth.The experiment was divided into three groups: blank control group(the essence with no containing extract),positive control group(5% minoxidil tincture)and experimental group(the essence with containing extract,group A).Before the hair grows,the speed of the back turned to black was observed through the naked eye;After the hair grows,the length of the hair was measured to determine the growth rate of the hair;The number of follicles was counted on 7th and 18 th and the follicular morphology wasobserved by microscopy on 18 th.The study found: hair grows before,the group A and positive control group turned to black fastly;After the hair grows,the A group and the positive control group had a similar growth rate,but the length of hair was the longest in the positive control group,and there were significant difference compared with the blank control group(P < 0.01);The number of hair follicles in mice was observed by microscope,and the number in the positive control group and A group was higher than that in blank group on 7th and 18 th.On 18 th,the results showed compared with the blank control group,in the positive control group and A group the hair follicles were more dense,surrounded by the dermal papilla,there were a lot of melanin in the hair bulb,hair follicles are still in the anagen.The results showed the essence can speed up the growth of mouse hair and extend the anagen and increase the number of hair follicles,to promote hair growth,it indicates that the extract had the same effect.30 volunteers who match the test requirements were selected for human efficacy test.Appropriate amount of essence was applied in the designated parts on scalp every day,by detecting the amount of sebum,counting the number of hair loss,counting hair follicle number in the unit area,calculating the hair growth rate and hair loss area.84 days later,the volunteers of the test indicators had improved,The specific manifestations: sebum secretion decreased,the number and area of hair loss decreased,the number of hair follicles increased,etc.The results show that the anti-hair loss essence can effectively improve the hair loss symptoms of patients with AGA.In summary,the extract can inhibit 5α-reductase activity effectively,inhibit the growth of microorganisms that cause inflammation on the scalp,and increase the microcirculation of the skin.And the essence containing the extract can improve the hair loss symptoms of patients with AGA.
