Effects Of Tangke Decoction On MiR-192 Signaling Pathway In Diabetic Nephropathy Rats

ObjectiveTo study the protective effect of Tangke Decoction on kidney of diabetic nephropathy rats,and to explore the potential mechanism of its therapeutic effect.Methods60 male SD rats,and 10 were randomly selected as normal control group.The remaining rats were injected with streptozotocin 30mg/kg in tail vein to establish the diabetic mellitus rat models.Diabetic mellitus rats were randomly divided into model group,Tangke Decoction prevention group(4.5g/kg),low dose group(4.5g/kg),high dose group(18g/kg)and Fosinopril sodium group(0.9mg/kg).Each group was administered by dosing.Blood glucose,24 h urinary protein,body weight and left kidney weight were measured.Kidney weight/body weight is calculated.Hematoxylin-eosin staining was used to observe the pathological changes of renal tissue.The expression of mi R-192 was detected by real-time fluorescence quantitative PCR.The m RNA expression of TGF-β1,SIP1 and Col1a2 were detected by real-time fluorescence quantitative PCR.The protein expression of TGF-β1,SIP1 and Collagen I in kidneys were detected by Western-blot.ResultsCompared with the model group,the 24 h urinary protein,left kidney weight and kidney weight/body weight were significantly decreased in the TangkeDecoction prevention group,low dose group,high dose group and Fosinopril sodium group(P<0.01);glomerular hypertrophy,basement membrane thickening,mesangial cell proliferation,extracellular matrix and other pathological damage was improved;the m RNA expression of TGF-β1 and Col1a2 in renal tissue were significantly decreased(P<0.05);the expression of mi R-192 in renal tissue was significantly decreased(P<0.05);the m RNA and protein expression of SIP1 was significantly increased(P<0.05);the protein expression of TGF-β1 and Collagen I in renal tissue was significantly decreased(P<0.05).The treatment effect of Tangke Decoction high dose group was better than that of other treatment groups.ConclusionsTangke Decoction can treat the kidney of diabetic nephropathy rats.The mechanism of treatment may be associated with reducing the expression of TGF-β1,mi R-192,Col1a2,Collagen I and enhancing the expression of SIP1.The treatment effect of Tangke Decoction high dose group was better.
