ObjectiveTo investigate the effect and mechanism of acupuncture combined with Tangke apozem on renal tissue in rats with diabetic nephropathy,and to study the renal fibrosis and oxidation in rats with type 2 diabetic nephropathy by acupuncture combined with the treatment of Tangke apozem.The influence of action and mechanism of action provide a theoretical basis for the treatment of patients with clinical diabetic nephropathy.MethodsA total of 54 SPF-class SD male rats were randomly selected from the blank control group,and the remaining rats were subjected to diabetic nephropathy(DN)model replication.After 4 weeks of high-fat and high-sugar diet,the rats were injected with the streptozotocin(30 mg/kg)in the tail vein,and the 24-hour urine protein(≥30 mg)was measured after 2 weeks.Divided into model group,irbesartan group(13.5mg/kg),acupuncture combined with Tangke apozem group(Acupuncture combined with traditional Chinese medicine group;18g/kg),Tangke apozem treatment group(Chinese medicine group;18g/kg),Acupuncture group.Oserve the general condition of the rats,and measure body weight(measured every seven days),blood sugar,kidney weight/body weight,24-hour urine protein,urea nitrogen(BUN),serum creatinine(Scr),triglyceride(TG)and total cholesterol(TC);HE staining,Masson staining and glycogen staining(PAS)were used to observe changes in renal tissue morphology;The serum levels of total superoxide dismutase(T-SOD),glutathione peroxidase(GSH-PX)and malondialdehyde(MDA)were measured by ELISA;Western-blot method was used to detect the protein expression levels of TGF-β1,MMP9,Collagen I(Collagen I),Collagen III(Collagen III),CTGF,and TIMP1 in kidney tissues.ResultsCompared with the traditional Chinese medicine treatment group and the model group,the kidney weight/body weight,24 h proteinuria,serum TG,BUN,Scr and TC levels in the Acupuncture combined with traditional Chinese medicine treatment group were significantly lower(P<0.05),The blood glucose level decreased significantly(P<0.05),the acupuncture combined with traditional Chinese medicine treatment can increase the activity of T-SOD and GSH-PX in rats(P<0.05),and effectively inhibit MDA production(P<0.05),and slow tissue cell damage.In addition to the acupuncture group,the other treatment groups significantly inhibited the expression of TGF-β1,CTGF and TIMP1 in rat kidney tissue,increased the expression of MMP9 protein in renal tissues(P<0.01),and accelerated the hydrolysis of Collagen I and Collagen III(P<0.05).Compared with traditional Chinese medicine treatment,the Acupuncture combined with traditional Chinese medicine treatment can improve the pathological morphology of renal tissue,slow down glomerular fibrosis,and reduce serum TG,BUN,Scr,TC,MDA content(P<0.05),in renal tissue.The expressions of TGF-β1,CTGF,TIMP1,Collagen I and Collagen III were significantly decreased(P<0.05),and the activities of T-SOD and GSH-PX in serum and the expression of MMP9 protein in renal tissues were increased.ConclusionsAcupuncture combined with Tangke apozem can effectively improve the blood sugar,blood lipids,24 h proteinuria,BUN,Scr and TC levels in diabetic nephropathy rats to delay renal function in rats with diabetic nephropathy;improve the anti-oxidative repair ability of rat kidney tissue,Acupuncture combined with Tangke apozem can regulate TGF-β1/CTGF signaling pathway by inhibiting TGF-β1/TIMP1/MMP9,reduce ECM deposition,and delay renal fibrosis in diabetic nephropathy rats. |
