Objective:Through the observation of Dahuangfuzi Decoction in treating chronic renal failure(CRF)rat,renal function,renal pathological changes and the effect of PI3K/Akt signal pathway,and explore its mechanism in the treatment of CRF.Method:①The modeling and grouping:60 at the age of 7 weeks of clean grade,weighing between 180-220g,Wistar rats,randomly divided into five groups with 12 rats in each group were named as blank group,sham operation group,model group,control group and treatment group.The model group,control group and treatment group of rats 5/6 with renal resection,mining way to establish animal model,adaptive feeding a week after the first surgery,anesthesia in chloral hydrate resection of left renal 2/3 in rat renal tissue.One week after the second surgery,resection of the right kidney of rats;rats in sham group were only removed two kidney capsule;blank group did not.②The treatment medicine:after feeding for 4 weeks,every morning rats in each group were given different drugs(10ml/Kg·d):blank group,model group and sham operation group rats using distilled water;the control group using Niaoduqing solution gavage;treatment group with supplemented Dahuangfuzi Decoction.Continuous gavage for 8 weeks..③The detection index:after 8 weeks treatment,the venous blood was collected under anesthesia by abdominal aorta of rats,for detecting blood routine,renal function;extraction the rat’s kidney,the residual renal tissue segmentation into two parts,one part of the renal tissue in the freezing tube,in order to use phosphate Western blot assay tissue Akt(p-Akt),total Akt(Pan Akt)protein content,another part of the renal tissue stored in paraformaldehyde,renal tissue were observed by light microscope and images with pathological changes were compared after treatmentResult:①Model group,control group and treatment group compared with blank group and sham operation group.RBC,Hb content decreased significantly.SCr,BUN content increased significantly.The ratio of p-Akt/pan Akt in renal tissue were significantly increased.The difference was significant(P<0.05).Under the light microscope,renal interstitial fibrosis were observed②Control group and the treatment group compared with model group,RBC,Hb content increased.SCr,BUN significantly reduced.The ratio of p-Akt/pan Akt in renal tissue were significantly decreased.The difference was significant(P<0.05).Under light microscope,the degree of renal fibrosis was lower than that of model group.③The treatment group compared with the control group,RBC,HB content increased,Scr,BUN significantly reduced the ratio of p-Akt/pan,the ratio of Akt in renal tissue were significantly decreased.The difference was significant(P<0.05),under the light microscope,renal interstitial fibrosis reduced compared with model group.Conclusion:①The supplemented Dahuangfuzi Decoction can significantly improve renal function of CRF rats,correct anemia,delaying the process of renal fibrosis;②The mechanism of supplemented Dahuangfuzi Decoction in the treatment of CRF may be achieved through down-regulation of PI3K/Akt signaling pathway. |
