| Objective: A study explores mechanism action of Taohe Chengqi Decoction in delaying Chronic renal failure,which provides ideas and objective experimental evidence for treatment of Chronic renal failure by observing the influences of Taohe Chengqi Decoction on chronic renal failure rats’ blood routine,renal function,the form of renal pathology,and Axin on Wnt/β-catenin signal path.Methods:Taking model of 5/6 nephrectomy chronic renal failure rats as object,125 aged 7 weeks adult male Wistar rats were fed for a week,after that,they were divided into several groups as follows,group of blank,sham operation,model,uremic clearance and Taohe Chengqi Decoction.The group of model,uremic clearance,and Taohe Chengqi Decoction adopted 5/6 nephrectomy copy CRF model,under the anesthesia of choral hydrate,2/3 of the rats’ left renal tissues were excised.A week later,the second operation was performed,the rats’ right renal tissues were excised and the two renal tunica of the rats in sham operation group were stripped,while the blank group did not be disposed.Three days after operation,the penicillin would be injected into enterocoelia to prevent infection.After the completion of modeling,the rats in each group were fed for four weeks,four weeks later,they were given different drug therapy at 9 am everyday.The rats in blank group,sham operation group and model group was given gavage with normal saline.The rats in uremic clearance group was given gavage with mix of uremic clearance granule and suspension.The rats in group of Taohe Chengqi Decoction was given gavage with Taohe Chengqi Decoction.After fifty-six days gavage persistently,the rats were sacrificed and their abdominal aorta blood were taken to detect RBC,Hb,Scr,BUN.Parts of their left renal tissue were placed in the cryopreserved tube and use Western Blot to detect the protein level of Wnt1、Axin、β-catenin in left renal tissue,in addition,detected the expression of Wnt1、Axin、β-catenin by real-time FQ-PCR.Another parts of renal tissue were put in stationary liquid.After treatment,pathologic conditions of renal tissue in each group were observed with light microscope and pictures were collected to compare the variation.Results:(1)Compared with the model group,the content of RBC and Hb in the Taohe Chengqi Decoction group had varying degrees increased.(P<0.05).(2)The results that serum SCr and BUN of Taohe Chengqi Decoction group were lower than those of model group and uremic clearance group(P<0.05)have statistically significance.(3)The results of Western Blot showed that compared with the model group,the protein expressions of Wnt1 and β-catenin in the left renal tissue were significantly decreased in Taohe Chengqi Decoction group(P<0.05),and the Axin expression level was up-regulated(P<0.05).(4)Real-time PCR revealed that compared the model group with uremic clearance group,the expressions of Wnt1 and β-catenin were down-regulated(P<0.05)and the expressions of Axin were up-regulated(P<0.05).(5)Under light microscope,HE staining of renal tissue showed that left renal tissue structure in the blank group and the sham operation group were normal.In the model group,the left renal tissue structure was disordered,glomerular edema and hyperplasia of renal interstitial fibers,and inflammatory cell infiltration was observed.Other than that,compared with uremic clearance group and model group,the degree of renal fibrosis was obviously improved in Taohe Chengqi Decoction.Conclusion:(1)Taohe Chengqi Decoction can improve renal anemia condition of CRF rats effectively by resolving stasis and turbidity and facilitating bowel movement,beyond that,it can lower CRF rats’ serum creatinine,urea nitrogen content and alleviate the renal pathology injury of rats.(2)In the 5/6 nephrotomy rats model,Taohe Chengqi Decoction can slow down the growth of renal fibrosis to delay the progression of chronic renal failure by inhibiting the Wnt1/β-catenin signal,down-regulating the expression of Wnt1,β-catenin and up-regulating that of Axin. |
