The Anti-depressive Mechanism Of Chaihu-guizhi Decoction On Depressive Mice

Objective:Observe the effect of Chaihu-guizhi decoction on the depressive behavior and the monoamine neurotransmitters of depressive mice model,and throught it to probe the anti-depressive mechanism of Chaihu-guizhi decoction.Mithods:1. The tests of tail suspension(TST), forced swimming(FST) and reserpine experiment were carried out to estimate that Chaihu-guizhi decoction has anti-depressant effect2. Compared with the depression in the control group,through the weight measurement,sugar water preference experiment to evaluate building success in mice.3. Detectation about the monoamine neurotransmitters of the mice brains: Chronic unpredictable stress were applied successively to set up depressive mice model.High performance liquid-electrochemical determination of concrete was applied to detect the monoamine neurotransmitters(5-HT、DAa nd NE) of the brains.Results:1. Compared with the depression in the control group, high-dose and middle-dose of Chaihu-guizhi decoction shortened the dead time of tail suspension and forced swimming in mice and resisted the descent of body temperature 、eyelid and body movement.2. Compared with the depression in the control group,high-dose and middledose of Chaihu-guizhi decoction increased the body quality,body weight in mice and the percentage of sugar water preference.3. Compared with the depression in the control group,high-dose and middledose of Chaihu-guizhi decoction highered the content of Single amineneurotransmitter 5-HT, DA and NE.Conclusion:Chaihu-guizhi decoction had anti-depressant effect and rel-i eved the depressant behaviors of mice. The mechanism mainly was due to i ncreasing the cerebral 5-HT、DA、NE and gastrointestinal hormone concentr ation in the brains.
