Objective: Based on the establishment of hepatocarcinoma rat model carrying abnormal savda syndrome, to investigate abnormal savda munziq’s effect on Hepatocarcinoma rat model carrying abnormal savda syndrome and its mechanism. Methods: On the basis of Uygur traditional medicine theory, establishing the abnormal savda syndrome rat model,future to including hepatocarcinoma of carrying abnormal savda syndrome rat model using Diethy(DEN) and administrated with abnormal savda munziq low, medium and high does group respectively. The serum protein were analyzed by proteo Miner low abundance protein enrichment system combined by(iTRAQ) technology, To establish differential expression profile of serum protein,(IPA) online bioinformatics software was applied to the function analysis of candidate serum proteins including the functional Annotation, regulatory network, biological function, canonical pathway and biomarker, which had been used to find out the potential biomarker for abnormal savda rat model with hepatocarcinoma and the formulas of potential targets for research. Results:The proteomics analysis identified 29 protein differentially expressed in serum of abnormal savda rat with hepatocarcinoma in response to treatment with the prescription. Bioinformatics analysis shows that these proteins are mainly involved in the pathogenesis of liver cancer related blood coagulation reaction, inflammation reaction, etc. These differentially expressed proteins mainly involved in retinol X receptor(RXR) pathway; By further application of IPA software of biomarkers screening, we found that found ORMI,NAP1L4,KNG1,TF,RAB27 B maybe targeted by its prescription. Conclusion: The Hepatocarcinoma carrying Abnormal savda rat model establishment is stable and reliable. The treatment of Abnormal Savda with its corresponding prescription,The abnormal savda munziq has an effect on whole regulatory network of protein expression, with ORMI,NAP1L4,KNG1,TF,RAB27 B as potential targets. |
