A.JRhizoma is one of perennial herb of Actractyca, belonging to composite family. In The Chinese Folk, A.J as a drug, which is widely used in treating stomach ulcers, chronic gastritis and other digestive diseases, and it’s very effective. It is mainly distributed in Northeast of China, Japan, Korea and other countries. It abounded in Heilongjiang and Jilin Province, and the eastern mountains of Liaoning Province.In order to make full use of the abundant wild resource on Changbai mountain and to promote A.J record into Pharmacopoeia of People’s Republic of China soonly, to research the chemical compound systematically.In this study, at first, the dry rhizome powder was extracted by CO2 supercritical fluid and the extrats were analyzed by GC-MS applications to identify the chemical composition of volatile oil of A. J;secondly, the remaining materia medica was extracted by 70% Athanol, and the Alcohol extracts were used to extract by ethylacetate and butanol extraction by turns. And then supercritical CO2 extract and each extration of alcohol extracts were further separated and make sure to gain the structure of the resulting compounds. To dry the remaining materia medica of accomplishment of Alcohol extraction on the sun, then, using the method of water extraction and alcohol precipitation to gain the crude polysaccharides, in final, using the phenol-sulfuric acid method to determine the content of the polysaccharide in its rude polysaccharides.Experimental results show that:(1) With CO2 supercritical fluid as solvent, the volatile oil constituents in A. J rhizomes were extracted, and the extracted substances were isolated and identified by GC-MS. The relative content of each constituent in volatile oil constituents were ascertained by peak area normalization of chromatogram. More than 40 kinds of chemical components were identified, which took up more than 94% of the total amount. Supercritical CO2 extraction was brown-yellow oily substance, which yield of 5.46%. (2)By using silica gel column chromatography, they were further separated and 6 compounds were isolated in supercritical CO2 extracts, ethyl acetate extracts and butanol extracts. Through the use of’H-NMR,13C-NMR and other spectroscopic data for structural dientification, they were identified as butenolideⅡ(I), β-sitosterol(Ⅱ), n-tetracontane(Ⅲ), butenolideⅢ(Ⅳ), n-hexadecanen (Ⅴ),β-daucosterol (Ⅵ). (3)The crude polysaccharides were extracted by water in A.J rhizome, using phenol-sulfuric acid method for edtermi nation of the content of polysaccharides in the crude polysaccharides, content of 55.49%, so polysaccharides of the A.J rhizome were at high levels.
