Effects Of Selective Cyclooxygenase-2Inhibitor NS-398on Proliferation And Invasion Of Human Cholangiocarcinoma Cell Line QBC939 | | Posted on:2013-07-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:W D Sun | Full Text:PDF | | GTID:2284330371473350 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective:To investigate the expression of Cox-2and VEGF in cholangiocarcinoma, analyze theirs relevance; To study the effect of selective Cox-2inhibitor (NS-398) on the proliferation and invasion in human cholangiocarcinoma cell QBC939.Methods:To detect the expression of Cox-2and VEGF in human cholangiocarcinoma by the Immunohistochemical method; QBC939cells were treated with NS-398at different concentrations, NS-398on the growth suppression in human cholangiocarcinoma cell line QBC939was detected by the MTT method; the invasive ability of NS-398on QBC939cells was detected by Transwell chamber.Results:High expression of Cox-2and VEGF in human cholangiocarcinoma tissues, there was significant difference between cholangiocarcinoma and normal tissues(P<0.01), and both were correlated (r=0.563, P<0.001); NS-398inhibited proliferation of the QBC939cells, with time and concentration increased, the inhibitory effect of NS-398on QBC939cells was more obviously; The maximum inhibitory concentration was100μmol/L. When the concentration greater than100μmol/L, the difference was not statistically significant (P>0.05) between72hours and48hours.
