Objective:To investigate the expression of Cox-2and VEGF in cholangiocarcinoma, analyze theirs relevance; To study the effect of selective Cox-2inhibitor (NS-398) on the proliferation and invasion in human cholangiocarcinoma cell QBC939.Methods:To detect the expression of Cox-2and VEGF in human cholangiocarcinoma by the Immunohistochemical method; QBC939cells were treated with NS-398at different concentrations, NS-398on the growth suppression in human cholangiocarcinoma cell line QBC939was detected by the MTT method; the invasive ability of NS-398on QBC939cells was detected by Transwell chamber.Results:High expression of Cox-2and VEGF in human cholangiocarcinoma tissues, there was significant difference between cholangiocarcinoma and normal tissues(P<0.01), and both were correlated (r=0.563, P<0.001); NS-398inhibited proliferation of the QBC939cells, with time and concentration increased, the inhibitory effect of NS-398on QBC939cells was more obviously; The maximum inhibitory concentration was100μmol/L. When the concentration greater than100μmol/L, the difference was not statistically significant (P>0.05) between72hours and48hours. QBC939cells were intervened by NS-398after36hours, with the concentration increased, the invasion ability was reduced significantly(P<0.01).Conclusion:Cox-2and VEGF express highly in cholangiocarcinoma, and both are correlated; NS-398inhibits the growth proliferation and decreases invasion ability of human cholangiocarcinoma QBC939cell in vitro.
