| Object: To evaluate the expressions of NGF and VEGF in hilar cholangiocarcinoma and their relationships with tumor's biological behaviour.Method:The specimens were collected from 45 patients with hilar cholangiocarcinoma in the first affiliated hospital of AnHui Medical University from May 2001 to May 2009. Among all, the ratio of male and female was 29:16; the age varied from 31 to 83 years old and the median age was 53; 30 cases had PNI and 15 ones without. Radical surgery were performed in all cases and lymphadenectomy were used. Additional, 20 bile duct specimens were carried from the Calculus of bile duct specimens as control. By using SP immunohistochemistry, we studied the expressions of NGF and VEGF in hilar cholangiocarcinoma and their relationships with tumor's biological behaviour. metastasis and prognosis. SPSS 13.0 was used to analyze the data.Result: 1. The expressions of NGF and VEGF in hilar cholangiocarcinoma were significantly higher than that in the control. Immunohistological staining with NGF and VEGF specific polyclonal antibodies can be found in cytoplasmic and membranous in the hilar cholangiocarcinoma cells.2. The expressions of NGF and the expressions of VEGF were significantly correlated.3. The positive expression of NGF in PNI group was higher than that in non-PNI group; The positive expression of VEGF in PNI group was no higher than that in non-PNI group; 4. By Kaplan-meier and Cox regression model analysis, NGF,VEGF and PNI lymph node metastasis were found to be independent parameter for prognosis. Also, TNM staging affected the rate of survival.Conclusions:1. NGF, VEGF may be secreted mainly by the bile duct cancer cells and expressed. And the expression of NGF can promote the expression of VEGF. The expressions of VEGF was closely related to the degree of malignancy of cholangiocarcinoma and also promotes bile duct along the blood vessels and lymphatic invasion and metastasis of hilar cholangiocarcinoma2. The expressions of NGF and the incidence of PNI were the close relationship but the expressions of VEGF was not associated with PNI. NGF, VEGF, and PNI , Lymph node metastasis affected the postoperative survival of the independence of the indicators.TNM staging had an impact on survival time of patients. |
