| Objective:To detect the plasma DNA colon cancer patients, the relationship between the pathological features of DNA in plasma in patients who have colon tumors,analysis on the early diagnosis, treatment and prognosis of colon tumor plasma DNA quantitative or qualitative value.Methods:We collected40colon cancer cases from2012November to2013January who received treatment,these patients met the clinical criteria for the diagnosis of colon tumor, patients with male27cases,female13cases, age between29-74years old, their average age was48.4±1.5years old. In the experiment, the medical staff on the patients collected venous blood, EDTA-K2anticoagulation, and under the centrifugal10min, and then to the plasma at4℃centrifugal10min. Then, using quantitative DNA detection in plasma by double fluorescent quantitative PCR containing, according to the result of detecting tumor staging, grading (according to the AJCC cancer staging manual (Sixth Edition)). The experimental set in40cases in the control group, for healthy people over the same period in our hospital physical examination,19male,21female, age30-88years old, their average age was47.4±1.6years old.Two groups of patients with age, hospitalization time data analysis indexes had no statistical significance (P>0.05).Results:In the experiment, two groups tests were successful, the experimental results showed:Patients with colon cancer (49.9)(30.2-61.3) plasma DNAcontent of ng/ml was higher than that of control group (healthy) was18.2(13.9-26.2) ng/ml (P<0.05). The plasma DNA content in patients with colorectal carcinoma with tumor staging and grading (P<0.05), and the content of patient age, gender, tumor size had no significant difference in plasma of patients with DNA (P>0.05), but the levels of plasma DNA in patients with advanced colon tumor, poor differentiation in which will be on the rise.In the experiment, using non parametric ROC analysis on the diagnostic efficacy of quantitative analysis of plasma DNA of patients with digestive tract cancer, area under the curve(AUC) and the diagnostic sensitivity is superior to the traditional indexes of serum CEA. Therefore, patients with tumors of digestive tract (especially the colon cancer patients), clinical patients on plasma DNA quantitative, qualitative detection of greater value.Conclusion:Clinical, for colon cancer patients, the plasma DNA was higher than that in the control group patients. It has a certain value for clinical diagnosis and treatment of colon cancer patients (that can be used as one of biochemical indexes diagnosed colon tumor). In addition, the plasma DNA level will be higher in patients with early colon cancer, it has important diagnostic value for malignant tumor of digestive tract. For tumor patients with undiagnosed, medical personnel should as soon as possible to detect the plasma DNA of the patients, to avoid missed diagnosis or misdiagnosis. |
