| Objective: Detecting the expression and mutation of the cycle-dependent proteinkinase8(CDK8) and the expression of beta-catenin in the colon cancer, studing thepossible mechanisms which induce colon cancer by CDK8,we analysised the relationshipbetween expression of CDK8and the features of clinicopathological for patients in coloncancer in order to provide some advise and the experimental basis for the moleculartreatment of colon cancer.Methods:48patients diagnosised of colon cancer patients in which take the coloncancer tumors and adjacent normal tissues where margins far2cm away from cancertumors.(1)Real time-PCR tested the expression of CDK8, β-catenin in tumor tissue andadjacent normal tissues, and the correlation and differences between them were analysised.(2) immunohistochemical testing the expression of CDK8, β-catenin in tumor tissue andadjacent normal tissues, the correlation and differences between them were analysised, wealso analysised the correlation clinical with pathological features.(3) the dideoxysequencing to test the sequencing cDNA of CDK8in colon cancer tissue,and analysisedCDK8gene mutation location, frequency, and the mutations and the relationship betweenthe clinical and pathological feature.Results:(1) we found CDK8, β-catenin gene expression in tumor tissue was higherthan the adjacent tissues by real time-PCR (all p<0.05), and the expression of CDK8,β-catenin in the tumor tissue with cancer and adjacent tissues had positive correlation.(2)immunohistochemical was found that:â‘ the expression of CDK8, β-catenin in tumortissue was higher than in adjacent tissues (all p<0.05),â‘¡CDK8, β-catenin protein intumor tissue and adjacent tissues in. expression was positively correlated;â‘¢and the expression of CDK8contact with TNM stage in patients. the expression positive rate ofCDK8in0, â… and â…¢ patients were significantly higher than the positive rate of Phaseâ…¡ â…£ patients, the difference was statistically significant (p<0.05);â‘£the positive rate ofCDK8with lymph node metastasis was significantly higher than that without lymph nodemetastasis, the difference was statistically significant (p<0.05);⑤there were nocorrelation between CDK8expression and tumor differentiation (p>0.05).(3) the dideoxysequencing assay analysis the colon cancer tissue found that CDK8-cDNA had a numberof mutations point and there were no significant correlation with the pathological features(all p>0.05).Conclusion: The expression of CDK8, β-catenin in the tumor tissue in colon cancerand adjacent tissues positive correlation; there has a certain relationship the expression ofCDK8and TNM stage; there were a large number of mutations in CDK8-cDNA of tumortissue and adjacent tissues; Therefore, CDK8is likely to change through mutation andexpression of the impact of β-catenin expression and thus likely caused cancer. |
