| Purpose: Studying on Yang Jiedu Tongluo Decoction for the treatment of essentialhypertension in rats, to explore Qianyang Jiedu Tongluo Decoction mechanism from theChinese out "wind phlegm blood stasis collaterals" pathogenesis point of treatment ofhypertensive rats.Method: Healthy male12weeks old male spontaneously hypertension rat (SHR)70,10WKY rats, randomly divided into Yang Group, detoxification group, Tongluo group, thewhole Party group, valsartan group, treatment group, SHR control group and normal controlgroup. Respectively before and after administration of2,4,6,8weeks with rat tail arterynon-invasive blood pressure measuring instrument Coda20208(the United States KentScientific Corporation) were measured in conscious quiet tail arterial systolic pressure (SBP),fixed time in the morning9:00-12:00. Each mouse continuous measurements of SBP3,1-2min interval, average. After2months of administration, kill the rats and take theirabdominal aorta and left ventricles. Measure the concentration of serum HS-CRP, TNF-a,IL-1β by using the ELISA kit. Observation of myocardial cells arranged in HE staining,observe the change of collagen content under the light microscope, Masson staining.Expression of PPAR-γ, TNF-a and TLR4RT-PCR mRNA in myocardial cells of the rats.Result: Qianyang Jiedu Tongluo Decoction (full set) can effectively inhibit theexpression of SHR in rat serum IL-1β, TNF-α and hs-CRP inflammatory factors, withstatistical significance (P<0.05). At the gene level, in the SHR rat myocardial tissue, the Partygroup and Yang group after treatment, the expression of TNF-a was significantly lower thanthe other treatment group and positive control (P<0.05), valsartan group and positive controlgroup than the expression of pioglitazone and other groups by the significant difference(P<0.05), and and the whole Party group and Yang group similar trend; in the expression ofTLR4, there were significant differences in Yang Group and the rosiglitazone group comparedwith the other treatments and the control group (P<0.05); the expression of PPAR-γ, thewhole group was significantly higher than that of the other groups. Histologically, the wholeParty group, valsartan group and pathological pioglitazone group of myocardial cells andmyocardial tissue in a regular arrangement, close, and other treatment group and controlgroup had significant difference.Conclusion: Confirm the treatment of primary hypertension rats from the "wind phlegmblood stasis collaterals" pathogenesis angle Qianyang Jiedu Tongluo Decoction (SHR) iseffective, its mechanism may be related to effects on PPAR-γ way, improve the expression ofinflammatory factors related to. |
